摘 要:目的建立如达-6中木香烃内酯和去氢木香内酯的含量测定方法。方法采用高效液相色谱法,Chrom Core 120-C_(18)(4.6×250 mm,5μm)色谱柱,甲醇-水(65:35)为流动相;流速1.0 ml/min;柱温30℃;进样量为20μL;检测波长为225 nm。结果木香烃内酯和去氢木香内酯分别在0.399-7.984μg(r=0.9993)、0.527-10.536μg(r=0.9999)范围内呈现良好的线性关系。对供试品进行精密度试验结果显示,木香烃内酯和去氢木香烃内酯的RSD分别为0.57%<和0.89%。对供试品进行稳定性试验结果显示,木香烃内酯和去氢木香内酯的RSD分别为1.02%<和1.13%。木香烃内酯和去氢木香烃内酯的平均加样回收率分别为98.12%、98.53%,二者的RSD分别为1.80%、1.42%。结论用高效液相色谱法测定如达-6中木香烃内酯和去氢木香内酯的含量具有专属性、精密度、稳定性良好等优点。Objective To establish a method for the determination of costunolide and dehydrocostone in Ruda-6.Methods HPLC was performed on Chrom Core 120-C_(18) column(4.6 ×250 mm,5μm) with methanol-water(65:35) as mobile phase.Flow rate 1.0 mL/min;Column temperature 30 ℃.Sample size:20 μL;The detection wavelength was 225 nm.Results The linear ranges of costunolide and dehydrocostolide were0.399~7.984 μg(r=0.9993) and 0.527~10.536(r=0.9999),respectively.The precision test results showed that the RSDS of costunolide and dehydrocostunolide were 0.57% and 0.89%,respectively.The stability test results showed that the RSD of costunolide and dehydrocostone were 1.02% and 1.13%,respectively.The average recoveries of costunolide and dehydrocostunolide were 98.12% and 98.53%,with RSDS of1.80% and 1.42%,respectively.Conclusion HPLC method for the determination of costunolide and dehydrocostlactone in Ruda-6 has the advantages of specificity,precision and good stability.
