摘 要:目的观察大黄素对大鼠角膜碱烧伤后炎症因子表达及瘢痕形成的影响,并分析作用机制。方法构建角膜碱烧伤大鼠模型,选取构建成功的64只大鼠随机分为模型组、大黄素低剂量组、大黄素中剂量组、大黄素高剂量组,每组16只,各大鼠左眼健康角膜为正常对照组。大黄素低、中、高剂量组大鼠右眼分别予浓度为10、20、40 mg/kg体质量的大黄素50滋l滴眼,正常对照组和模型组用1%DMSO溶液50滋l滴眼,4次/d。给药第3、7、14、28天每组4只大鼠用裂隙灯检查角膜混浊度。实验开始后第7天每组处死12只大鼠获取角膜组织样本,检测角膜组织病理学改变并进行炎症细胞计数,ELISA法检测角膜组织中炎症因子水平,免疫荧光法检测角膜组织Ⅳ型胶原(ColⅣ)、琢平滑肌肌动蛋白(琢-SMA)表达情况,Western blot法检测角膜组织转化生长因子-茁1(TGF-茁1)、碱性成纤维细胞生长因子(b-FGF)表达情况。结果与模型组比较,实验开始后第3、7、14、28天大黄素低、中、高剂量组角膜浑浊度评分均降低(均P<0.05),且大黄素高剂量组低于大黄素低剂量组(均P<0.05);角膜组织炎症细胞计数及IL-2、TNF-琢、IFN-酌水平均降低(均P<0.05),且大黄素低剂量组>大黄素中剂量组>大黄素高剂量组(均P<0.05);角膜组织ColⅣ、琢-SMA免疫荧光累积光密度值及TGF-茁1、b-FGF蛋白相对表达水平均降低(均P<0.05),且大黄素低剂量组>大黄素中剂量组>大黄素高剂量组(均P<0.05)。结论大黄素可有效抑制大鼠角膜碱烧伤后炎症反应及瘢痕形成,可能通过抑制炎症因子及TGF-茁1、b-FGF表达发挥作用。Objective To investigate the effect of emodin on scar formation and the expression of inflammatory factors in rat corneas after alkali burn,and the related mechanism.Methods The cornea burn injury model was induced by alkali treatment on the right eyes in rats.Sixty four rats with cornea burn were randomly divided into model group,low-,medium-,and high-dose emodin group with 16 rats in each group,and the healthy corneas on the left eyes of rats were used as the control group.The low-,medium-and high dose emodin groups were given 50μl of 10,20,40 mg/kg emodin for local instillation,and the control group and model group were given 50μl of 1%DMSO solution instead,4 times a day.Corneal turbidity was checked by slit lamp on d3,d7,d14,and d28 after administration.On the 7th day of the administration,12 rats in each group were sacrificed to obtain corneal tissue samples,the histopathological changes of the cornea were detected and the inflammatory cells were counted.The inflammatory factors in corneal tissue was detected by ELISA.The expressions of corneal tissue type IV collagen(Col IV)andα-smooth muscle actin(α-SMA)were detected by immunofluorescence method.The expressions of ransforming growth factor-β1(TGF-β1)and basic fibroblast growth factor(b-FGF)in corneal tissues were detected by Western blot.Results Compared with the model group,the scores of corneal turbidity in the low-,medium-,and high-dose emodin groups were decreased at d3,d7,d14,and d28(all P<0.05),and the scores of high-dose emodin group were lower than those of the low-dose emodin group(all P<0.05).Compared with the model group,the inflammatory cell counts and the interleukin-2(IL-2),tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ)levels in the corneal tissue of the low-,medium-,and high-dose emodin groups were decreased(all P<0.05),and the degree of decrease was in the order of high-dose group,medium-dose group and low-dose group(all P<0.05).Compared with the model group,the ColIV,α-SMA immunofluorescence IOD value and the relative expres
