摘  要:在黄瓜幼苗茎段石蜡切片制作的教学实验中,以常规石蜡切片方法为基础,通过延长材料固定时间、70%低浓度乙醇脱水时间以及脱蜡和复水时间,缩短高浓度乙醇脱水时间和番红染色时间,改良浸蜡、包埋、展片、烘片、封片方式,得到了连续、完整的蜡带,使最终获得的切片在显微观察下,可见茎横切面结构完整、染色清晰。优化后的石蜡切片技术,避免了常规制片过程中出现的组织变硬变脆、组织与蜡带分离、组织硬化导致切片碎裂、蜡带断裂、组织脱片、固绿颜色过深覆盖番红染色等问题。Based on the conventional paraffin section method in the teaching experiment of paraffin sectioning of cucumber stems,the material fixation time,dehydration time of low-concentration of 70%ethanol,and dewaxing and rehydration time are extended,and the dehydration time of high-concentration ethanol and safranin staining time are shortened,and wax dipping,embedding,display,baking,and sealing methods are improved.Continuous and complete wax tape can be obtained based on the improved paraffin section technique,and it can be seen that the cross-sectional structure of the stem is complete and the staining is clear through microscopic observation.The optimized paraffin section technology can avoid the problems of the hardening and brittleness of the tissue,separation of the tissue and wax band,and hardening of the tissue leading to section fragmentation,wax band breakage,tissue peeling off,and excessively deep coverage of the solid green color in the conventional sectioning process.
