摘 要:目的探讨5-氧代-6,8,11,14-二十碳四烯酸(5-oxo-6,8,11,14-eicosatetraenoic acid,5-oxo-ETE)体外能否促进鼻息肉中嗜中性粒细胞(neutrophils,Neu)释放炎症介质髓过氧化物酶(myeloperoxidase,MPO)。方法用免疫组织化学法检测正常鼻黏膜组织和鼻息肉组织中Neu的浸润,将两种组织进行体外培养,用酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)法检测其中MPO含量,后将5-oxo-ETE或5-oxo-ETE+百日咳毒素(pertussis toxin,PT)加入两种组织的培养基中,再以ELISA法检测其中MPO浓度。结果鼻息肉组织中Neu数目明显多于正常鼻黏膜组织,体外培养的鼻息肉组织中MPO含量显著高于正常鼻黏膜组织,单纯5-oxo-ETE干预后鼻息肉中MPO浓度明显升高,而5-oxo-ETE+PT干预后该炎症介质浓度下降,但与未经任何干预的鼻息肉中MPO含量相比,无统计学差异。结论5-oxo-ETE体外可以促进鼻息肉组织中Neu释放炎症介质MPO。OBJECTIVE To explore whether 5-oxo-6,8,11,14-eicosatetraenoic acid(5-oxo-ETE)promoted the release of myeloperoxidase(MPO)from neutrophils(Neu)in nasal polyps(NP)in vitro.METHODS We used immunohistochemistry method to detect the infiltration of Neu in normal nasal mucosa and NP tissues.Then these two tissues were cultured in vitro,and the concentration of MPO was examined in them with enzyme linked immunosorbent assay(ELISA).After that,5-oxo-ETE or 5-oxo-ETE+pertussis toxin(PT)were administered into the cultures,and MPO in them was evaluated using ELISA again.RESULTS The number of Neu was increased in NP compared to normal nasal mucosa.The concentration of MPO was also up-regulated in NP cultured in vitro compared with that in normal tissue.After with the intervention of 5-oxo-ETE,MPO was enhanced greatly in NP,however,the level of MPO was reduced significantly after 5-oxo-ETE+PT administration.There was no statistical difference between 5-oxo-ETE+PT intervention and NP tissue.CONCLUSION 5-Oxo-ETE promotes the release of MPO from neutrophils in NP in vitro.
