摘 要:为了研究氧化型低密度脂蛋白受体1(oxidized low density lipoprotein receptor,OLR1)基因的多态性及其不同基因型对肉用性状的影响,试验以58头草原红牛为研究对象,采用PCR扩增和Sanger测序方法检测草原红牛OLR1基因第4外显子的遗传多态性,利用"牛等家养动物群体遗传分析工具V1.0"评价草原红牛群体遗传结构,利用SPSS 19.0软件中One-Way ANOVA程序分析ORL1基因不同基因型对肉用性状的影响。结果表明:OLR1基因在草原红牛中存在多态性,PCR扩增片段大小为390 bp,与预期片段大小一致,且ORL1基因外显子4在Chr5:99 812 807 bp处存在A>G突变,该基因有AA、GG和AG 3种基因型,AA和A分别为优势基因型和优势等位基因,且A/G位点的基因分布符合Hardy-Weinberg平衡(P>0.05),AA基因型对失水率的影响显著高于AG型(P<0.05),GG基因型对眼肌面积的影响显著高于AG型(P<0.05)。说明ORL1基因的多态位点有可能成为草原红牛肉用性状选育的分子标记。In order to investigate the polymorphism of oxidized low density lipoprotein receptor( OLR1) gene and the influence of different genotypes on meat traits,58 red steppe cattle were selected as research subjects,genetic polymorphism in OLR1 gene exon four of grassland red cattle was detected by PCR amplification and Sanger sequencing,the genetic structure of Grassland red cattle population was evaluated by“Cattle and other domestic animal population genetic analysis tool V1. 0”,and the effects of different ORL1 gene genotypes on meat traits were analyzed by One-Way ANOVA program in SPSS 19. 0 software. The rsults showed that OLR1 gene was polymorphic in Grassland red cattle,the PCR amplification fragment was 390 bp,which was consistent with the expected fragment size,and exon 4 of ORL1 gene had A/G mutation at Chr5:99 812 807 bp. There were three genotypes,AA,GG and AG. AA and A were dominant genotype and allele,respectively,and the gene distribution of A/G locus was Hardy-Weinberg equilibrium( P>0. 05). The effect of AA genotype on water loss rate was significantly higher than that of AG genotype( P< 0. 05),and the effect of GG genotype on eye muscle area was significantly higher than that of AG genotype( P<0. 05). These results indicated that the polymorphic loci of OLR1 gene might be a molecular marker for trait selection of Grassland red cattle.
