摘 要:为了确定引起某警犬基地42只犬发生趾间炎的病原菌,试验对患犬进行临床检查,并从患犬病健组织交界部位采集病料,分别通过革兰氏染色镜检、细菌分离培养、生化鉴定、PCR诊断、药敏试验对病原菌进行检测。结果表明:患犬脚趾肿大,部分犬在脚趾及周围出现结节、脓疱、脚趾间皮肤坏死呈煤焦油样,解剖死亡犬可见胰腺部分坏死。92.86%的患犬可在病变部位检测到坏死梭杆菌,50.00%的患犬可在病变部位检测到金黄色葡萄球菌,30.95%的患犬可在病变部位检测到化脓性链球菌。采集的病料在血琼脂培养基上培养16~18 h后,出现灰黄色、表面光滑、中间略凸起、边缘不整齐的菌落,且菌落周围出现明显溶血环,革兰氏染色镜检可见丝状、长短不一的革兰氏阴性杆菌。生化鉴定结果与坏死梭杆菌阳性对照株一致。PCR扩增得到大小约为349 bp的目的片段,与坏死梭杆菌参考株同源性在99%以上;坏死梭杆菌对林可霉素、头孢唑啉、氨苄西林、多西环素高度敏感,对青霉素、头孢哌酮钠、卡那霉素、阿莫西林中度敏感,对阿米卡星、链霉素、庆大霉素、四环素耐药。说明引起此次群发性趾间炎的主要病原菌为坏死梭杆菌,该菌可以转移到胰腺并导致胰腺坏死,进而引发犬的急性死亡。In order to determine the pathogen causing the interdigititis in 42 dogs of a police dog base,the test carried out clinical examinations on the affected dogs,and collected disease materials from the junctions of the diseased and healthy tissues of the affected dogs.The pathogenic bacteria were detected through microscopic examination,bacterial isolation and culture,biochemical identification,PCR diagnosis,and drug susceptibility test.The results showed that the toes of the affected dogs were swollen,and some dogs had nodules,pustules on and around the toes;the skin necrosis between the toes was coal tar-like,and the necrosis in part of the pancreas could be seen in the dead dog after anatomy.92.86%of the affected dogs could detect Fusobacterium necrophorum at the lesion,50.00%of the affected dogs could detect Staphylococcus aureus at the lesion,and 30.95%of the affected dogs could detect Streptococcus pyogenes at the lesion.After the collected disease materials were cultured on blood agar medium for 16 to 18 hours,colonies with grayish-yellow,smooth surface,slightly raised middle and irregular edges appeared,and obvious hemolysis loops appeared around the colonies.Gram staining microscopic examination showed that filamentous,gram-negative bacilli of varying lengths.The biochemical identification results were consistent with the positive control strain of Fusobacterium necrophorum.PCR amplification yielded a target fragment with a size of about 349 bp,which had a homology of more than 99%with the reference strain of Fusobacterium necrophorum.Fusobacterium necrophorum was highly sensitive to lincomycin,cefazolin,ampicillin,doxycycline,and moderately sensitive to penicillin,cefoperazone sodium,kanamycin,amoxicillin,and resistant to amikacin,streptomycin,gentamicin and tetracycline.The results suggested that the main pathogen causing this mass interdigititis was Fusobacterium necrophorum,which could transfer to the pancreas and cause pancreatic necrosis,and then lead to acute death in dogs.
