摘  要:为了研究乌苏里貉资源群体中GHR基因的单核苷酸多态性(SNP),试验选取生长性状差异较大的两个乌苏里貉群体分别构建DNA池,对GHR基因的第10外显子进行PCR扩增并测序。结果表明:发现5个SNP位点,分别为T911C、C1112T、G1454A、G1522A、C2279T。其中T911C为错义突变,可导致苯丙氨酸改变为亮氨酸;C1112T为错义突变,可导致丝氨酸改变为苯丙氨酸;G1454A为同义突变,氨基酸不发生改变;G1522A为错义突变,可导致精氨酸改变为赖氨酸;C2279T为错义突变,可导致丝氨酸改变为亮氨酸。表明这4个错义突变的SNP位点有可能与乌苏里貉生长性状相关。In order to study the single nucleotide polymorphisms(SNP) of GHR gene in Wusuli raccoon dogs resource population, two Wusuli raccoon dog groups with significant differences in growth traits were selected to construct DNA pools respectively. The exon 10 of GHR gene was amplified by PCR and sequenced. The results showed that five SNPs were found, which were T911 C, C1112 T, G1454 A, G1522 A and C2279 T. Among them, T911 C is a missense mutation, which can cause a change from phenylalanine to leucine. C1112 T is a missense mutation that can change serine to phenylalanine. G1454 A is the synonymous mutation and amino acid does not change. G1522 A is a missense mutation that can change arginine to lysine. C2279 T is a missense mutation that can change serine to leucine. It indicated that the SNP loci of these four missense mutations might be related to the growth traits of Wusuli raccoon dog.
