摘 要:目的研究清热中药对2型糖尿病小鼠代谢及氧化应激水平的影响及作用机制。方法研究以雄性小鼠为研究对象构建2型糖尿病模型,基于传统的黄连-生地黄中药对,将地骨皮和鬼箭羽两项具有抗氧化应激活性成分的传统清热药物与其相结合构成清热药物组方,通过测量小鼠血糖和氧化应激关键指标(SOD和MDA)的变化情况,分析该组方对于小鼠糖尿病及其并发症的效果。结果与对照组比较,模型组和治疗组小鼠体质量增长减缓,血糖浓度、胰岛素浓度和MDA均表现显著升高,而抗氧化应激指标SOD显著下降,出现多饮多尿等典型2型糖尿病症状。采用黄连-生地黄组方和黄连-生地黄-地骨皮和鬼箭羽组方对治疗组小鼠进行处理后,其血糖浓度、胰岛素浓度和MDA出现一定的回调,而SOD则出现一定的上升。结论黄连-生地黄-地骨皮和鬼箭羽组方具有更好的效果和潜在临床应用价值。Objective To study the effect and mechanism of heat-clearing Chinese herbs on metabolism and oxidative stress levels in type 2 diabetic mice. Methods In this study, male mice were used as the research object to construct a type 2 diabetes model. Based on the traditional Chinese medicine pair of Huanglian(Coptidis Rhizoma) and Shengdihuang(Rehmanniae Radix), the two traditional heat-clearing medicines with anti-oxidative stress active ingredients were compared with Digupi(Lycii Cortex) and Guijianyu(Euonymus alatus). Combined with a prescription for clearing away heat, the effects of this prescription on diabetes and its complications in mice were analyzed by measuring the changes in blood glucose and key indicators of oxidative stress(SOD and MDA) in mice. Results Compared with the control group, the body mass growth of the model group and the treatment group slowed down, blood glucose concentration, insulin concentration and MDA were significantly increased, while the anti-oxidative stress index SOD was decreased significantly, and polydipsia and polyuria were typical symptoms of type diabetes. After treating mice in the treatment group with Huanglian(Coptidis Rhizoma)-Shengdihuang(Rehmanniae Radix)prescriptions and Huanglian(Coptidis Rhizoma)-Shengdihuang(Rehmanniae Radix)-Digupi(Lycii Cortex)-Guijianyu(Euonymus alatus) prescriptions, the blood glucose concentration, insulin concentration and MDA of the mice in the treatment group showed a certain callback, while the SOD showed a certain increase. Conclusion The formulae of Huanglian(Coptidis Rhizoma)-Shengdihuang(Rehmanniae Radix)-Digupi(Lycii Cortex)-Guijianyu(Euonymus alatus) have better effects and potential clinical application value.
