摘 要:目的为探究灸法对机体免疫功能良性调整作用的核心作用因素,从程序性死亡受体(PD-1)及免疫细胞分子的角度,观察不同灸法对环磷酰胺所致免疫抑制兔PD-1、PD-L1及CD19、CD45R、CD69、NKG2D、SP的影响。方法40只大耳白兔随机分为空白组、免疫抑制模型组、隔药饼灸组以及艾灸组。除空白组外,后3组均以环磷酰胺60 mg/kg量进行腹腔注射,连续7 d,造成免疫抑制模型。次日隔药饼灸组、艾灸组分别进行干预治疗,隔日施灸,共10次。治疗结束第2天抽取静脉血,分离血清。结果造模后,与空白组比较,模型组实验前后重差值降低(P<0.01),模型组PD-1、PD-L1含量显著升高(P<0.01,P<0.05);CD19、NKG2D含量明显降低(P<0.01),CD45R含量明显升高(P<0.05);艾灸干预后,两组实验前后体质量差值均明显升高(P<0.01),PD-1、PD-L1含量明显降低(P<0.01,P<0.05);艾灸组和隔药饼灸组CD19、NKG2D含量均明显升高(P<0.05),隔药饼灸组CD45R含量明显降低(P<0.01);CD69、SP组间比较差异无统计学意义(P>0.05)。结论隔药饼灸与艾灸对免疫抑制兔实验前后体质量、PD-1、PD-L1及免疫细胞分子CD19、CD45R及NKG2D干预作用显著,两种灸法在对PD-L1及CD45R的影响中呈现差异。Objective In order to explore the core factors of moxibustion on the benign adjustment of immune function,the effects of different moxibustion on PD-1,PD-L1,CD19,CD45 R,CD69,NKG2 D and SP in immunosuppressive rabbits induced by cyclophosphamide were observed from the perspective of PD-1 and immune cell molecules.Methods Forty rabbits were randomly divided into the blank group,the immunosuppressive model group,the hebaral cake-separated moxibustion group and the moxibustion group.Except the blank group,other groups were injected intraperitoneally with cyclophosphamide 60 mg/kg for 7 consecutive days,resulting in an immunosuppressive model.The next day,the hebaral cake-separated moxibustion group and the moxibustion group were given intervention treatment,performed every other day for a total of 10 times.When the treatment was finished,the venous blood was taken out and the serum was separated.Results For the model group,the difference in body weight before and after the experiment was reduced(P<0.01).Compared with those of the blank group,the contents of PD-1 and PD-L1 of the model group were significantly increased(P<0.01 or P<0.05).Meanwhile,the contents of CD19 and NKG2 D were significantly reduced(P<0.01)and the content of CD45 R was significantly increased(P<0.05)in the model group.After the moxibustion intervention,the body weight difference between the two groups was significantly increased(P<0.01)and the contents of PD-1 and PD-L1 were decreased(P<0.01,P<0.05).The contents of CD19 and NKG2 D in the moxibustion group and the hebaral-separated cake moxibustion group were significantly increased(P<0.05).The content of CD45 R in the hebaral cake-separated moxibustion group was significantly reduced obviously(P<0.01).There were no significant differences of CD69 and the SP among the groups(P>0.05).Conclusion Two moxibustion methods have significant effects on body weight,PD-1,PD-L1,CD19,CD45 R and NKG2 D in immunosuppressed rabbits.The two moxibustion methods have different effects on PD-L1 and CD45 R.
