摘 要:目的研究苍术炒焦鞣质含量变化及对脾虚泄泻证的药理药效,探明焦苍术长于"固肠止泻"的物质基础及作用机制。方法采用干酪素法测定苍术炒焦前后鞣质含量,并建立脾虚泄泻动物模型,研究焦苍术鞣质对模型大鼠急性腹泻、消化道炎症、结肠对水分吸收能力及脑肠神经系统的调控作用。结果焦苍术鞣质含量增加125%以上;鞣质给药组腹泻指数与TNF-α极显著降低(P<0.01),IL-10极显著升高(P<0.01),结肠AQP_(3)水平显著增高(P<0.05),结肠切片HE染色炎症明显转归,5-HT及5-HT4R、5-HT7R表达水平极显著降低(P<0.01)。结论鞣质是焦苍术"固肠止泻"主要物质基础之一,其通过抑制消化道炎症、促进结肠对消化道水分的吸收、抑制5-HT及5-HT4R、5-HT7R过高表达,恢复肠道正常的吸收和蠕动功能。Objective To study the changes of tannin content of deep-fired Cangzhu(Rhizoma Atractylodis)and its pharmacological effects on syndrome of spleen deficiency and diarrhea and explore the material basis and mechanism of Cangzhu(Rhizoma Atractylodis).Methods The tannin content of Cangzhu(Rhizoma Atractylodis)was determined by casein method before and after deep-fired.In addition,an animal model of spleen deficiency and diarrhea was established to study the effects of tannin of Cangzhu(Rhizoma Atractylodis)on acute diarrhea,digestive tract inflammation,colonic water absorption capacity and cerebral and intestinal nervous system in rats.Results The tannin content of deep-fired Cangzhu(Rhizoma Atractylodis)increased by 125%.In the tannin-treated group,diarrhea index and content of TNF-αwere significantly reduced(P<0.01).The content of IL-10 was significantly increased(P<0.01),and the level of AQP_(3)in the colon was significantly increased(P<0.05).The inflammation was significantly reduced in HE staining of colon slices,and the expression levels of 5-HT,5-HT4 R and 5-HT7 R were significantly reduced(P<0.01).Conclusion Tannin of deep-fired Cangzhu(Rhizoma Atractylodis)is one of the main material basis of its"intestinal fixation and diarrhea prevention",which can restore normal intestinal absorption and peristolic function by inhibiting digestive tract inflammation,promoting colon’s absorption of digestive tract water,and inhibiting overexpression of 5-HT,5-HT4 R and 5-HT7 R.
