摘 要:目的研究健脾益肠散对2,4,6-三硝基苯磺酸(TNBS)诱导的溃疡性结肠炎(UC)模型大鼠共刺激分子CD80、淋巴细胞功能相关抗原-1(LFA-1)表达的影响。方法SD大鼠随机分为正常组、模型组、柳氮磺吡啶组和健脾益肠散高、中、低剂量组,采用TNBS溶液灌肠法建立UC大鼠模型,给药21 d后取材,免疫组织化学法检测结肠组织中CD80、LFA-1的蛋白阳性表达,RT-PCR法检测结肠组织中CD80、LFA-1的mRNA表达。结果模型组大鼠结肠组织中CD80的蛋白阳性和mRNA表达均明显增加,LFA-1的蛋白阳性和mRNA表达均明显减少,与正常组比较差异有统计学意义(P<0.01);健脾益肠散高、中、低剂量均可不同程度地降低CD80的蛋白阳性和mRNA表达、升高LFA-1的蛋白阳性和mRNA表达,与模型组比较差异有统计学意义(P<0.05或P<0.01);且健脾益肠散高剂量组优于健脾益肠散中、低剂量组(P<0.05或P<0.01)。结论健脾益肠散对UC肠黏膜的免疫炎症抑制作用可能与调节结肠组织中CD80、LFA-1的表达有关。Objective To investigate the effect of Jianpi Yichang Powder(健脾益肠散,JYP)on the expressions of costimulatory molecules CD80 and lymphocyte function-related antigen-1(LFA-1)in a rat model of 2,4,6-tritonitrobenzene sulfonic acid(TNBS)-induced ulcerative colitis(UC).Methods SD rats were randomly divided into normal group,model group,sulfasalazine group and JYP high,medium and low dose groups.The UC rat model was established by TNBS solution enema.After 21 days of administration,the positive expressions of CD80 and LFA-1 protein in colon tissue were detected by immunohistochemistry,and the CD80 and LFA-1 mRNA expressions in colon tissue were detected by Real-Time PCR.Results The protein positive and mRNA expressions of CD80 in colon tissues of rats in the model group were significantly increased,LFA-1 protein positive and mRNA expressions were significantly decreased,and there was statistical difference compared with those of the normal group(P<0.01).The high,middle and low doses of JYP could reduce the CD80 protein positive and mRNA expressions,increase the LFA-1 protein positive and mRNA expressions,which was statistically different from that of the model group(P<0.05 or P<0.01).And the high dose group of JYP was better than that of medium and low dose groups(P<0.05 or P<0.01).Conclusion The immunosuppressive effect of JYP on UC intestinal mucosa may be related to the regulation of CD80 and LFA-1 expressions in colon tissue.
