摘 要:目的汤剂是一种存在着多种分散相态的液态分散体系,其中胶体相态粒径为纳米级,也称纳米相态。为探究汤剂中纳米相态是否客观存在,选择白虎汤为研究对象,对其相态进行分离并表征,为解析传统汤剂的科学性提供方法学参考。方法通过透射电镜观察白虎汤纳米相态,并对透射电镜支持网(超薄碳膜和微栅)、相态拆分方法(葡聚糖凝胶、火棉胶膜以及透析袋)、拆分次数(1次、2次、3次、4次、5次)以及纳米相态保存时间(0、8、24 h)进行考察,同时采用马尔文粒径仪及透射电镜对所获得的纳米相态进行表征。结果纳米相态分离所用的透射电镜支持网应选择超薄碳膜,分离方法采用透析袋进行透析,最佳拆分次数为4次,保存时间以0 h为宜。分离得到的白虎汤纳米粒子形态呈球形,粒径大小主要集中在100 nm左右,zeta电位为-3.11 mV,含有C、O、Si、Na、Ca、Mn、Fe、Cu、Na等元素,其中铁元素含量较高。该相态不稳定,易聚集。结论纳米相态在白虎汤中客观存在,并可采用现代研究方法进行表征。Objective Decoction is a kind of liquid dispersion system with many dispersed phases.The particle size of colloidal phase is nanometer,also called nanometer phase.In order to explore the existence of nano phase in decoction,this paper selected Baihu Decoction(白虎汤)solution for phase separation,obtained the best conditions for the separation of nano phase and characterized the nano phase,so as to provide methodological reference for the analysis of the scientificity of traditional decoction.Methods The nanophase state of Baihu Decoction was observed by transmission electron microscope,and the transmission electron microscopy support network(ultra thin carbon film and micro grid),phase splitting method(dextran gel,collodion film and dialysis bag),resolution times(1,2,3,4,5 times)and nano phase preservation time(0,8,24 h)were investigated.At the same time,the nano phase was characterized by Malvin particle size meter and transmission electron microscope.Results Ultra thin carbon membrane should be selected as the transmission electron microscope support network for nano phase separation.The separation method adopted dialysis bag for dialysis.The best separation times was four times and the storage time was 0 h.The isolated Baihu Decoction nanoparticles were spherical in shape,the particle size was mainly about 100 nm,and the zeta potential was-3.11 mV.They contained elements such as C,O,Si,Na,CA,Mn,Fe,Cu and Na,among which the content of iron was high.The phase state was unstable and easy to gather.Conclusion Nano phase exists in Baihu Decoction objectively and can be characterized by modern research methods.
