行气健脾方对动脉粥样硬化家兔心肌细胞线粒体凋亡相关蛋白表达影响研究

摘　　要：目的探讨行气健脾方对动脉粥样硬化(AS)家兔心肌细胞线粒体凋亡的影响。方法选取清洁级雄性家兔18只,随机分为对照组、模型组、西药组及行气健脾方低、中、高浓度组,各3只。模型组、西药组及行气健脾方低、中、高浓度组用高脂饲料饲喂法建立AS模型,对照组家兔喂以标准饲料。模型建立后,模型组及对照组分别予以0.9%氯化钠10 mL·kg^(-1),每日1次灌胃;西药组予以0.15 mg·mL^(-1)瑞舒伐他汀溶液10 mL·kg^(-1),每日1次灌胃;行气健脾方低、中、高浓度组分别予以0.35 g·mL^(-1)、0.7 g·mL^(-1)、1.4 g·mL^(-1)行气健脾方10 mL·kg^(-1),每日1次灌胃。所有家兔均连续给药10周。检测比较各组家兔血脂水平,心肌细胞凋亡指数,以及心肌组织细胞色素C(Cytochrome C)、分裂的含半胱氨酸的天冬氨酸蛋白水解酶3(cl-caspase3)、B淋巴细胞瘤-2(Bcl-2)、B细胞淋巴瘤-xl(Bcl-xl)、Bcl-2相关X蛋白(Bax)、Bcl-2相关k蛋白(Bak)表达水平。结果与模型组比较,西药组及行气健脾方低、中、高浓度组家兔血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)水平较低,高密度脂蛋白胆固醇(HDL-C)水平较高,心肌细胞凋亡指数较低,心肌组织Cytochrome C、cl-caspase3、Bax、Bak表达水平较低,Bcl-2、Bcl-xl表达水平较高,其中行气健脾方高浓度组改变最为显著,差异有统计学意义(P<0.05,P<0.01)。结论行气健脾方能浓度依赖性的调节AS家兔血脂水平,抑制心肌细胞凋亡,其作用可能与下调线粒体凋亡途径相关因子Cytochrome C、cl-caspase3、Bax、Bak表达,以及上调Bcl-2、Bcl-xl表达有关。Objective To investigate the effect of Xingqi Jianpi Recipe(行气健脾方)on mitochondrial apoptosis of myocardial cells in rabbits with atherosclerosis(AS).Methods 36 clean male rabbits were selected,and were randomly divided into blank control group,model group,western medicine group and Xingqi Jianpi Recipe low,medium and high concentration groups,each with 3 rabbits.Rabbits in model group,rosuvastatin western medicine group and Xingqi Jianpi Recipe low,medium and high concentration groups were established.AS models were set up by high-fat feeding.Rabbits in control group were fed with common pellet feed.After the models were established,the model group and blank control group were given 0.9%NaCl 10 mL·kg^(-1) respectively,intragastric administration once a day;rosuvastatin western medicine group was given 0.15 mg·mL^(-1)rosuvastatin solution 10 mL·kg^(-1),intragastric administration once a day;Xingqi Jianpi Recipe low concentration group,middle concentration group,high concentration group were given 0.35 g·mL^(-1),0.7 g·mL^(-1),1.4 g·mL^(-1) Xingqi Jianpi Recipe(10 mL·kg^(-1))respectively,intragastric administration once a day.All rabbits were administered continuously for 10 weeks.The serum lipid levels,cardiomyocyte apoptosis indexes,expression levels of Cytochrome C,cleaved caspase 3(cl-caspase 3),B-lymphoma-2(Bcl-2),B-cell lymphoma-xl(Bcl-xl),Bcl-2-related X protein(Bax),Bcl-2-related K protein(Bak)in myocardial tissue were detected and compared.Results Compared with model group,serum total cholesterol(TC),triglyceride(TG)and low density lipoprotein cholesterol(LDL-C)levels were lower,cardiomyocyte apoptosis indexes were lower,the expression levels of Cytochrome C,cl-caspase3,Bax,Bak in myocardium were lower,and the expression levels of Bcl-2,Bcl-xl were higher in rabbits of rosuvastatin western medicine group and Xingqi Jianpi Recipe low,medium and high concentration groups,and among them,the Xingqi Jianpi Recipe high concentration group had the most significant change,the differences were stati
