摘  要:目的尚未发现梅毒螺旋体(TP)对青霉素耐药,但临床上仍存在较高比例的经过青霉素规范治疗后未愈的梅毒血清固定患者。病原菌青霉素结合蛋白的表达水平或结构改变可导致病原菌对抗生素耐受性差异。分析TP的青霉素结合蛋白基因特征,可为揭示该基因在梅毒血清固定现象中的作用机制提供理论基础。方法使用生物信息学软件诠释TP全基因组ORF,对编码蛋白进行在线分析。结果梅毒螺旋体亚种Nichols株全基因组序列不含β-内酰胺酶基因,但含有3个编码青霉素结合蛋白mrcA、pbp1、pbp2基因,全长分别为2655 bp、1875 bp、1959 bp。mrcA、PBP1及PBP2均含有1个跨膜结构。mrcA含有信号肽,但PBP1和PBP2无信号肽。结论TP缺乏编码β-内酰胺酶基因可能是其对青霉素高度敏感的原因之一,但可通过提高青霉素结合蛋白基因表达量或基因结构改变来提高对青霉素的耐受性。Objective It has not been found that Treponema pallidum is resistant to penicillin,but there are still a high proportion of syphilitic sero-resistant patients who have not been cured after penicillin treatment.The change of expression level or structure of penicillin binding proteins of pathogenic bacteria can lead to the difference of antibiotic tolerance.The analysis of the gene characteristics of penicillin binding protein of Treponema pallidum can provide a theoretical basis for revealing the mechanism of the gene in the sero-resistance of syphilis.Methods The ORF gene of the whole genome of Treponema pallidum was interpreted by bioinformatics software,and the encoded protein was analyzed online.Results There was noβ-lactamase gene in the whole genome of Nichols,but there were three penicillin binding protein genes(mrcA,pbp1,pbp2),with a total length of 2655 bp,1875 bp,and 1959 bp respectively.mrcA,pbp1 andpbp2 contain one transmembrane structure respectively.mrcA contains a signal peptide,butpbp1 andpbp2 have no signal peptide.Conclusion The lack ofβ-lactamase gene in Treponema pallidum may be one of the reasons that Treponema pallidum is highly sensitive to penicillin.The results are helpful to reveal the mechanism of syphilitic sero-resistance,but need to be confirmed by scientific experiments.
