摘  要:为明确小菜蛾Plutella xylostella体内主要化感受体的表达分布特性,通过对特定组织转录组数据分析并结合半定量RT-PCR测定,研究小菜蛾气味受体(odorant receptor,OR)、离子型受体(ionotropic receptor,IR)和味觉受体(gustatory receptor,GR)3类化感受体基因在不同虫态、组织及性别中的表达特异性。转录组数据分析结果显示,共有48个OR、8个IR和15个GR基因在被测组织中表达,不同基因呈现不同的组织表达特异性。通过整合基因组和转录组数据分析共获得62个GR基因,其中48个为全长序列;系统进化树分析结果表明,48个全长GR中3个属于CO2受体,10个属于糖受体,35个属于苦味受体;成虫头(去除触角后)、触角、足、翅、雌成虫产卵器及4龄幼虫头部分别表达19、22、27、21、24和21个GR基因;成虫性别间比较发现,PxylGR2、PxylGR20、PxylGR61基因在雌成虫头部中表达量较高,PxylGR5、PxylGR23、PxylGR34、PxylGR37基因在雌成虫触角中表达量较高,PxylGR37、PxylGR53、PxylGR54基因在雌成虫足中表达量较高,而PxylGR17、PxylGR21和PxylGR22基因在雄性翅中表达量较高。In order to investigate the expression and distribution characteristics of the main chemoreceptors in diamondback moth Plutella xylostella,three chemoreceptors including odorant receptor(OR),ionotropic receptor(IR)and gustatory receptor(GR)in different developmental stages,tissues and sexes of P.xylostella were analyzed by semi-quantitative RT-PCR and transcriptome sequencing.The results of transcriptomic data showed that 48 ORs,eight IRs and 15 GRs were detected with different expression levels in different tissues.In addition,62 GR genes including 48 full-length genes were identified by analyzing genomic and transcriptomic data;based on phylogenetic analysis,three of these 48 genes belonged to CO2 receptor group,ten to sugar receptor group and 35 to bitter receptor group.Finally,semi-quantitative PCR analysis was carried out to determine the gene expression specificity in larval heads and different male and female tissues.The results showed that,among the 62 GR genes,21 were expressed in the larval heads,19 in the adult heads,22 in adult antennae,27 in adult legs,21 in adult wings,and 24 in female ovipositors.In particular,PxylGR2,PxylGR20,and PxylGR61 were female biased in expression in adult heads;PxylGR5,PxylGR23,PxylGR34 and PxylGR37 were female biased in adult antennae;PxylGR37,PxylGR53 and PxylGR54 were female biased in adult legs,while PxylGR17,PxylGR21 and PxylGR22 were male biased in adult wings.
