摘 要:为明确自然条件下养猪发酵床垫料对大肠杆菌生长的影响,探究大肠杆菌(Escherichia coli)在垫料中的生长状况,采用人工接种方法,将绿色荧光蛋白(GFP)标记的大肠杆菌K_(88)接种到养猪微生物发酵床垫料中;采用平板分离培养方法,观察大肠杆菌K_(88)-GFP(Escherichia coli K_(88)-GFP)在垫料中的生长动态。结果表明:E.coli K_(88)-GFP在灭菌处理的新鲜垫料和使用1年的旧垫料中均能很好生长,分离培养168 h含量分别达到4.03×10^(8) cfu·g^(-1)和5.37×10^(8) cfu·g^(-1),在数量级上无显著差别;而在未灭菌处理的新鲜垫料和使用1年旧垫料中均不能生长。说明在自然环境下,养猪微生物发酵床垫料对大肠杆菌K_(88)的生长有抑制作用。In order to clarify the effect of padding on the growth of Escherichia coli in the fermentation bed for pig raising under natural conditions,the growth of Escherichia coli in padding was studied.The green fluorescent protein(GFP)-labeled Escherichia coli K_(88) was inoculated into the padding of microbial fermentation bed for pig raising by using the method of artificial inoculation.The growth dynamics of Escherichia coli K_(88)-GFP in the padding were observed by using the plate isolating culture method.The results showed that Escherichia coli K_(88)-GFP could grow well both in the fresh padding with bactericidal treatment and the old padding used for one year,and the contents of Escherichia coli K_(88)-GFP reached 4.03×10^(8) cfu·g^(-1) and 5.37×10^(8) cfu·g^(-1) after 168 h of isolating culture,respectively,with no significant difference in order of magnitudes.However,Escherichia coli K_(88)-GFP could not grow in the fresh padding without bactericidal treatment and the old padding used for one year.The results indicated that the padding of microbial fermentation bed for pig raising could inhibit the growth of Escherichia coli K_(88) in the natural environment.
