摘 要:为揭示绵羊繁殖力的影响机制,利用分子标记辅助选择来快速提高绵羊的繁殖性能,本实验采用直接测序法对杜泊羊与湖羊群体5个基因中的突变位点进行了比较分析。结合测序后序列通过DNAStar软件预测蛋白质二级结构;统计基因型频率、基因频率、纯合度、杂合度和多态信息含量并将基因多态性与产羔数进行关联分析。结果表明:杜泊羊ESR2基因中的RS428438934位点处发生T→C突变,导致氨基酸发生了改变,CC与CT、TT与CT基因型之间产羔数差异显著;RS418841713在杜泊羊与湖羊中都具有多态性,处于不平衡状态;RS398521635、RS402413016、G7、G8、RS417060437、RS604746425在杜泊羊与湖羊中均只有1种基因型,无突变类型,处于Hardy-Weinberg平衡状态。In order to reveal the influence mechanism of sheep's fecundity and use molecular marker-assisted selection to quickly improve sheep's reproductive performance,this experiment used direct sequencing method to compare and analyze the mutation sites in 5 genes of Dorper sheep and Hu sheep population.Combined with the sequence after sequencing,the protein secondary structure is predicted by DNAStar software;the genotype frequency,gene frequency,homozygosity,heterozygosity and polymorphism information content are counted,and the genetic polymorphism is correlated with litter size.The results showed that the T→C mutation in the RS428438934 locus in the ESR2 gene of Dorper sheep resulted in changes in amino acids.There was a significant difference in litter size between CC and CT,and TT and CT genotypes;RS418841713 Both Dorper sheep and Hu sheep have polymorphisms and are in an unbalanced state;RS398521635,RS402413016,G7,G8,RS417060437,RS604746425 have only one genotype in both Dorper sheep and Hu sheep,and there is no mutation type,and they are in Hardy-Weinberg balance state.
