摘 要:本文旨在通过对种公鸡睾丸和附睾进行转录组测序和生物信息学分析,筛选出2种组织中的差异表达基因并分析其生物学功能,探究睾丸和附睾在鸡精子发生和成熟过程中的作用。本研究共以8只公鸡的睾丸和附睾组织为对象,采用RNA-seq技术分析2种组织中的基因表达情况,筛选出差异表达基因并对差异表达基因进行GO和KEGG通路富集分析。结果表明,在种公鸡的睾丸和附睾组织的样本中平均获得3.1×107条clean reads,其中80%的clean reads比对到鸡参考基因组上。与附睾组织相比,睾丸中共有5124个差异表达基因,包括2300个上调基因和2814个睾丸下调基因。GO富集分析表明,差异表达基因主要富集在细胞分化、细胞发育和细胞粘附以及蛋白质分解过程;KEGG通路主要富集在MAPK通路、谷胱甘肽代谢途径、细胞色素P450对外源物质的代谢作用途径、药物代谢-细胞色素P450途径。综合分析基因表达水平,GO和KEGG富集结果表明,睾丸中上调表达基因LAMTOR3、AVBD10、HPGDS可能在精子发生过程中发挥作用;附睾中上调基因TRIM36、TCP11在调节精子运动能力中发挥重要作用;睾丸在外源物质代谢中发挥重要作用。本研究为分析鸡睾丸和附睾功能差异提供了新的参考和理论依据。The aim of this study was to screen the differentially expressed genes in the testis and epididymis of breeder cocks by transcriptome sequencing and bioinformatics analysis,and to explore the role of testis and epididymis in the process of spermatogenesis and maturation.In this study,the testis and epididymis tissues of 8 cocks were used to analyze the gene expression in the two tissues by RNA-seq technique,and the differentially expressed genes were screened out and enriched by GO and KEGG pathways.The results showed that an average of 3.1×107 clean reads were obtained from testis and epididymis tissues of breeder cocks,and 80%of clean reads were compared to chicken reference genome.Compared with epididymis,there were 5124 differentially expressed genes in testis,including 2300 up-regulated genes and 2814 down-regulated genes.GO enrichment analysis showed that the differentially expressed genes were mainly concentrated in the process of cell differentiation,cell development,cell adhesion and protein decomposition;KEGG pathway was mainly concentrated in MAPK pathway,glutathione metabolism pathway,cytochrome P450 metabolic pathway on exogenous substances and drug metabolism cytochrome P450 pathway.Comprehensive analysis of gene expression level,GO and KEGG enrichment results showed that up-regulated genes LAMTOR3,AVBD10 and HPGDS in testis may play a role in spermatogenesis;up-regulated genes TRIM36 and TCP11 in epididymis play an important role in regulating sperm motility;testis plays an important role in exogenous material metabolism.This study provides a new reference and theoretical basis for analyzing the functional differences between the testis and epididymis in chickens.
