摘 要:目的研究PPAR-α激动剂非诺贝特对翼状胬肉成纤维细胞的抑制作用并初步探索其可能的机制。方法从翼状胬肉中分离得到成纤维细胞进行培养,给予非诺贝特后,测量细胞增殖和迁移的情况;运用IF、WB和RT-PCR来探究非诺贝特对其可能的作用机制。结果非诺贝特抑制了翼状胬肉成纤维细胞的增殖和迁移,并激活其受体PPAR-α,且下调了产生活性氧的关键酶NOX4及上调了抗氧化因子NRF2。并且,非诺贝特还降低了炎症因子IL-6和肿瘤坏死因子TNFα的表达。结论非诺贝特对翼状胬肉成纤维细胞具有抑制增殖和迁移以及抗炎、抗氧化的作用,这一作用有望用于翼状胬肉的治疗中。OBJECTIVE To investigated the inhibitory effects and mechanism of fenofibrate,an agonist of peroxisome proliferator-activated receptor alpha(PPAR-α)on the fibroblast cells of pterygium.METHODS The primary pterygium fibroblast cells were cultured and used in the experiment.The cell proliferation and migration were measured to examine the effects of fenofibrate.Immunofluorescent staining,western blot and quantitative real-time polymerase chain reaction assay were conducted to study the molecular mechanism of fenofibrate.RESULTS Fenof-ihrate significantly suppressed the cell proliferation and migration of cultured pterygium fibroblast cells.Fenofibrate specifically increased the level of PPAR-α.Fenofibrate inhibited the oxidative stress via blocking the ROS generating enzyme:NADPH oxidase 4(N0X4)and upregulating the antioxidant factor:NF-E2 related factor-2(NRF2).In addition,fenofibrate decreased the levels of inflammatory factors:interleukin 6(IL-6)and tumor necrosis factorα(TNFα).CONCLUSION Fenofibrate has anti-oxidant and anti-inflammatory activities on the fibroblast cells of pterygium.
