洛浦县双峰驼体表蚤形态学观察及分子鉴定

摘　　要：为鉴定新疆南疆双峰驼体表蚤的种类,从新疆洛浦县收集11峰家养双峰驼体表蚤67只,采用形态学观察初步鉴定种类后,基于蚤线粒体COII基因对蚤的DNA样本进行PCR扩增和种系发育分析。结果显示,67只蚤均为人蚤,其中雄蚤和雌蚤分别为23只和44只。随机选取8只蚤的DNA样本进行PCR扩增和测序,所获8只蚤的序列均与新疆地区虎鼬源人蚤的序列相似性100%。说明洛浦县双峰驼体表蚤的种类为人蚤,应加强其调查和监测工作。To identified the species of fleas in Bactrian camels in Southern Xinjiang.Sixty seven fleas were collected from 11 domestic Bactrian camels body surface in Lop county,and identified the species of fleas used by morphological observation,then fleas DNA samples were screened by PCR amplification and phylogenetic analysis based on fleas mitochondrial COII gene.The results showed that 67 fleas were all Pulex irritans by using morphological identification,with 23 males and 44 females.Eight flea DNA samples were randomly selected for PCR amplification and sequencing,and all the obtained sequences were identified as P.irritans,which had 100%homology with the isolates from a Vormela peregusna in Xinjiang.These findings revealed that the species of fleas was P.irritans from Bactrian camel body surface in Lop county,and the investigation and monitoring should be expanded in future.
