育成期高能饲喂下开产与未开产蛋鸡肝脏转录组差异分析

摘  要:对育成期高能饲喂的开产与未开产蛋鸡进行肝脏转录表达谱差异分析,探明在高能饮食状态下肝脏中影响开产的基因,为提高产蛋打下基础。以高能饲粮饲喂育成期蛋鸡为对象,运用高通量测序平台对开产与未开产鸡的肝脏进行转录组测序,使用HTSeq和DESeq等方法分析基因表达量并运用qRT-PCR验证,并以超几何分布对差异表达基因进行GO富集分析和KEGG富集分析。结果显示,各样本测序质量均较高。与未开产鸡相比,开产鸡有285个基因表达上调,209个基因表达下调。差异表达基因GO富集分析发现,有60个显著富集的GO功能(FDR<0.05),其中,44个富集在生物过程,11个富集在细胞组分,5个富集在分子功能。大部分富集的都是脂质代谢相关功能。差异表达基因KEGG富集分析发现1个显著富集的KEGG信号通路(FDR<0.05),即非不饱和脂肪酸生物合成,这是一个脂质代谢相关的信号通路,包含脂质代谢相关基因FADS1、FADS2、ACOT7、ACAA1和ELOVL6。GO和KEGG结果均表明脂质代谢对开产很重要。综上,育成期高能饲喂的蛋鸡能否提前开产,与肝脏脂质代谢过程及与其相关的一些基因表达水平有关。In this study,the liver transcriptional profiles of laying hens reaching and not reaching sexu-al maturity under high energy feed during growing period were analyzed to clarify the genes in liver-which can influence the sexual maturity under high energy feed.This study will lay a foundation forimproving egg production.High-energy feed was used to feed laying hens during growing period,andthe high throughput sequencing platform was used to sequence the transcriptome of the livers in layinghens reaching sexual maturity(R)and not reaching sexual maturity(N).The bioinformatics(HTSeqand DESeq)was used to analyze the gene expression levels and qRT-PCR was used to validate them.And the hypergeometric was used to analyze the differentially expressed genes(DEG)in GO andKEGG enrichment.The results showed that the sequencing qualities of all samples were good.Com-pared with the N group,the expression levels of 285 genes in the R group were up-regulated and 209genes were down-regulated.GO enrichment analysis of DEG showed that there were 60 significantlyenriched functions,among which 44 were enriched in biological processes,ll were enriched in cellularcomponents,and 5 were enriched in molecular functions(FDR≤0.05).Most of the enriched GO wereabout lipid metabolism.KEGG enrichment analysis of DEG showed that there was only one signifi-cantly enriched signaling pathway(Biosynthesis of unsaturated fatty acids)(FDR<0.05).It is relatedwith lipid metabolism,and containd lipid metabolic genes FADS1,FADS2,ACOT7,ACAAl andELOVL6.The GO and KEGG results revealed that lipid metabolism was important to sexual maturitty.In conclusion,whether laying hens under high energy feed during growing period reach sexual ma-turity early is related to lipid metabolic process and the expression levels of some lipid metabolic genesin their livers.
