摘 要:目的采用高效液相色谱连用紫外检测器检测经典名方半夏泻心汤中盐酸小檗碱的含量。方法色谱柱以十八烷基硅烷键合硅胶为填充剂,以乙腈0.05mol·L^(-1)及KH_(2)PO_(4)溶液(50∶50)为流动相,流速设置为1mL·min^(-1),检测波长为345nm。结果盐酸小檗碱出峰时间为9.5min左右,阴性样品对盐酸小檗碱的检测影响较小,试验方法有效。盐酸小檗碱在0.26~1.04μg之间呈良好线性关系,得回归方程为:Y=61.583X-0.9668,R^(2)=0.999。最终确定半夏泻心汤物质基准的盐酸小檗碱每剂含量范围33.12~61.50mg。结论该方法有效简便的测定出半夏泻心汤物质基准中盐酸小檗碱的含量,为经典名方半夏泻心汤的进一步研究与发展奠定了基础。OBJECTIVE The content of berberine hydrochloride in Banxia Xiexin Decoction was determined by HPLC with UV detector.METHODS The chromatographic column was filled with Octadecyl silane bonded silica gel,acetonitrile 0.05mol·L^(-1) and potassium dihydrogen phosphate solution(50∶50)as mobile phase,the flow rate was set to 1mL·min^(-1),and the detection wavelength was 345nm.RESULTS The peak time of berberine hydrochloride is about 9.5 minutes.The negative sample has little effect on the detection of berberine hydrochloride,and the test method is effective.Berberine hydrochloride showed a good linear relationship between 0.26~1.04μg,and the regression equation was:Y=61.583X-0.9668,R^(2)=0.999.Finally,the content range of berberine hydrochloride in each dose of Banxia Xiexin Decoction was determined to be 33.12~61.50mg.CONCLUSION This method is effective and simple to determine the content of berberine hydrochloride in the material standard of Banxia Xiexin Decoction,which lays a foundation for the further research and development of Banxia Xiexin Decoction.
