摘 要:从河北省保定市某养殖场采集疑似患病毒性腹泻/黏膜病(BVD)的犊牛粪便6份,进行牛病毒性腹泻病毒(BVDV)的分离培养,根据GenBank上登录的BVDV 5′-UTR片段设计引物,利用RT-PCR技术对目的片段进行扩增,扩增得到的目的基因片段送至公司测序,测序结果进行同源性分析和系统进化树构建。结果:分离出1株致细胞病变毒株,采用RT-PCR方法扩增出大小为288 bp的特异性条带,与预期大小相符,与GenBank中登录的部分BVDV毒株同源性为92.3%~99.0%,该毒株与T1-5(MN417828)、VR451(AY763031)、BJ175170(MT119454)、MF-1(KY865369)和MF-2(KY865370)在同一进化分支上,亲缘关系最近,将其命名为HB-BDZ。本研究为河北省内BVDV疫苗的制备及牛病毒性腹泻病的防控提供理论依据。Six feces samples were collected from cows suspected of suffering from bovine viral diarrhea/mucosal disease(BVDV)on a farm in Baoding City,Hebei Province.In order to isolate and identify the bovine viral diarrhea virus(BVDV),the samples were isolated and cultured.Primers were designed according to the BVDV 5′UTR gene logged on GenBank,and the methods of RT-PCR amplification technology,sequencing of target gene fragment,homology analysis and phylogenetic tree construction were used to identify and analyze the isolated viruses.The results showed that a cytopathic virus strain was isolated,and a specific band of 288 bp was amplified by RT-PCR method,which was consistent with the expected size,and had a homology of 92.3%-99.0%to some BVDV strains registered in GenBank.The strain was in the same branch and had the closest genetic relationship with T1-5(MN417828),VR451(AY763031),BJ175170(MT119454),MF-1(KY865369)and MF-2(KY865370),and it was named HB-BDZ.This study would lay a theoretical foundation for preparation of BVDV vaccine and for prevention and control of bovine viral diarrhea disease in Hebei Province.
