摘  要:目的:观察4周中等强度跑台运动对去卵巢大鼠抑郁样行为、海马炎症因子(IL-1β、IL-6、TNF-α)水平及海马CA1、CA3区nNOS表达影响,探讨运动改善去卵巢大鼠抑郁样行为的潜在机制。研究方法:将30只雌性成年大鼠随机分为三组,分别为假手术组(SHAM,n=10)、去卵巢组(OVX,n=10)、去卵巢运动组(OVX+EX,n=10)。运动结束后通过旷场实验及强迫游泳实验测试大鼠的抑郁样行为,采用ELISA法检测海马炎症因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)含量,利用免疫组化对海马CA1、CA3区nNOS阳性细胞个数及阳性产物面积进行测量。结果:与SHAM组相比,OVX组大鼠旷场实验中中央格次数、中央格时间百分比及总运动能力(跨格+直立次数)显著下降(P<0.05);强迫游泳实验中大鼠不动时间延长,攀爬时间显著下降(P<0.05);海马炎症因子(IL-1β、IL-6、TNF-α)水平显著增加(P<0.05);海马CA1区、CA3区nNOS免疫阳性细胞的数量与阳性细胞面积表达下降(P<0.05)。经过4周中等强度的跑台运动后,与OVX组比较,OVX+EX组大鼠旷场实验中中央格次数、中央格时间百分比及总运动能力(跨格+直立次数)增加(P<0.05);强迫游泳实验中大鼠不动时间减少,攀爬时间上升(P<0.05);海马炎症因子(IL-1β、IL-6、TNF-α)水平下降(P<0.05);海马CA1区、CA3区nNOS免疫阳性细胞的数量与阳性细胞面积表达增加(P<0.05)。结论:4周跑台运动可以改善去卵巢大鼠抑郁样行为,可能与此运动降低海马炎症细胞因子IL-1β、IL-6、TNF-α的释放、增强海马CA1、CA3区的nNOS表达有关。By observing the effects of 4-week moderate-intensity treadmill exercise on depression-like behavior,hippocampal inflammatory factors(IL-1β,IL-6,TNF-α)levels and the expression of nNOS in hippocampus CA1 and CA3 areas in ovariectomized rats,we aim to explore the potential mechanism for improving depressionlike behavior in ovariectomized rats by exercise.Thirty female adult rats were randomly divided into three groups:sham operation group(SHAM,n=10),ovariectomized group(OVX,n=10),ovariectomized exercise group(OVX+EX,n=10).After the exercise,the rats’depression-like behavior was tested by open field experiment and forced swimming experiment,and the content of hippocampal inflammatory factors of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)was detected by ELISA,the number of nNOS-positive cells and the area of positive products in hippocampal CA1 and CA3 areas are measured by immunohistochemistry.Compared with those of the SHAM group,the number of central grids,the percentage of central grid time and the total exercise capacity(cross grids+number of uprights)in the open field experiment of the OVX group were significantly decreased(P<0.05);in the forced swimming experiment,the immobile time of rats increased and the climbing time decreased significantly(P<0.05);hippocampal inflammatory factors(IL-1β,IL-6,TNF-α)levels significantly increased(P<0.05);the number of nNOS immunopositive cells and positive cell area expression in hippocampal CA1 and CA3 areas decreased(P<0.05).After 4-week moderate-intensity treadmill exercise,compared with those of the OVX group,the number of central grids,the percentage of central grid time and the total exercise capacity(cross grid+number of uprights)in the open field experiment of the OVX+EX group increased(P<0.05);in the forced swimming experiment,the immobility time of rats reduced and the climbing time increased(P<0.05);the levels of hippocampal inflammatory factors(IL-1β,IL-6,TNF-α)decreased(P<0.05);the number of nNOS immunopositive cells
