摘 要:目的总结近年来环氧合酶-2(COX-2)抑制剂的研究进展。方法通过归纳国内外COX-2抑制剂的研究进展报道,总结COX-2抑制剂与炎症相关疾病的作用机制,并按照化合物的结构类型进行分类,对具有显著体内外抗炎活性的化合物进行综述。结果化学合成和天然来源的COX-2抑制剂均具有良好的COX-2抑制作用,其中一些化合物采用计算机分子对接表现出与多个氨基酸形成氢键的特征,为其结构活性关系的论证提供了直接证据。结论选择性COX-2抑制剂有望成为治疗炎症相关疾病的潜在方法。Objective To summary the research progress of cyclooxygenase-2(COX-2)inhibitors in recent years.Methods Based on the current literature,the mechanisms of COX-2 inhibitors to treat inflammation-related diseases were summarized,and the compounds with significant anti-inflammatory activities were classified according to the chemical structure.Results Both synthesized and natural-derived COX-2 inhibitors had good COX-2 inhibitory effects.Some of the compounds showed the characteristics of forming hydrogen bonds with multiple amino acids by means of computer molecular docking,which provided direct evidence for the structure-activity relationship.Conclusion The selective COX-2 inhibitors are expected to become potential compounds for the treatment of inflammation-related diseases.
