摘  要:目的探讨桃红四物汤对不同程度兔软骨损伤模型Smad1信号转导蛋白表达的影响,从而阐明桃红四物汤干预Smad1信号转导蛋白表达促进软骨损伤修复作用机制。方法将40只健康新西兰大白兔,适应性喂养1周后,按照随机的方法平均分成四组,分别为桃红四物汤组、氨基葡萄糖组、模型组和正常对照组,每组各10只。对桃红四物汤组、氨基葡萄糖组和模型组进行软骨损伤造模,术后分别以桃红四物汤、氨基葡萄糖对对应组灌胃,模型组、正常对照组灌服等量生理盐水,并于灌胃后4周、6周两个时限,从每组随机抽取5只实验兔,麻醉后抽取腹主动脉血,并按原手术切口切开后暴露损伤软骨,进行大体观察,并切取包括股骨下端在内的关节面,检测Smad1信号转导蛋白表达情况,予以统计分析。结果桃红四物汤组平均血清Smad1浓度明显高于氨基葡萄糖组、模型组,但是低于正常对照组;WB检测的平均灰度值测定第4周时低于氨基葡萄糖组,但明显高于模型组,第6周时则高于氨基葡萄糖组;各组大白兔关节软骨组织HE染色病理观察,第4周与第6周差异不大。结论桃红四物汤能上调软骨损伤模型大白兔Smad1信号转导蛋白的表达,促进软骨损伤修复,并且上调效果优于氨基葡萄糖。Objective To explore the effect of Taohong Siwu Decoction on the expression of Smad1 signal transduction protein in rabbit model with cartilage injury of different degrees,so as to clarify the mechanism of Taohong Siwu decoction intervening the expression of Smad1 signal transduction protein and promoting cartilage injury repair.Methods A total of 40 healthy New Zealand white rabbits were randomly divided into 4 groups after adaptive feeding for 1 week,namely Taohong Siwu Decoction group,glucosamine group,model group and normal control group,with 10 rabbits in each group.The models with cartilage injury were made in Taohong Siwu Decoction group,glucosamine group and model group.After operation,Taohong Siwu Decoction and glucosamine were given to the corresponding groups respectively,and the model group and normal control group were given the same amount of normal saline.Five experimental rabbits were randomly selected from each group within four weeks and six weeks after gastric perfusion.After anesthesia,abdominal aorta blood was collected.The injured cartilage was exposed observed generally throng cutting the original surgical incision,and the articular surface,including the lower end of the femur,was incised,and the expression of Smad1 signal transduction protein was detected and statistically analyzed.Results The average concentration of serum Smad1 in Taohong Siwu Decoction group was significantly higher than that in glucosamine group and model group,but lower than that in normal control group.Western Blot(WB)detection showed that the average gray value in Taohong Siwu Decoction group was lower than that of glucosamine group at the 4 th week,but significantly higher than that of model group,and higher than that of glucosamine group at the 6 th week.Pathological observation of articular cartilage tissue of rabbits in each group by hematoxylin-eosin(HE)staining showed little difference between the 4 th week and 6 th week.Conclusion Taohong Siwu Decoction can up-regulate the expression of Smad1 signal transduct
