摘  要:目的探究复方木鸡颗粒中药效组分组成的各配伍组诱导肝癌HepG2细胞凋亡的药效学研究,筛选出药效组分最佳配伍比例,并将形成的新配伍复方与原复方进行药效差异性对比,从而较全面分析复方木鸡颗粒中药效组分诱导肝癌HepG2细胞凋亡的配伍规律,阐明其抗肝癌作用的药效物质基础。方法以复方木鸡颗粒6种药效组分为研究对象,设计并制作该微流控芯片,并结合均匀设计方法,以细胞凋亡坏死率为评价指标,开展药效组分最佳配伍比例研究,进而形成新的组分配伍复方。结果经灌注给药48 h后,复方木鸡颗粒各药效组分配伍组的细胞凋亡坏死率与空白组相比,均有显著性差异(P<0.05),且药效组分核桃楸皮黄酮,菟丝子黄酮,山豆根生物碱,山豆根多糖,菟丝子多糖,云芝多糖最佳配伍比例是4.35:5.80:15.03:10.73:1.36:1.20,并通过药效差异性对比试验,证明两者在相同剂量时,药效结果无显著性差异。结论本研究通过与微流控芯片技术的结合,初步揭示了复方木鸡颗粒药效组分抗肝肿瘤的组分组成规律,遵循中药组方理念,为抗肝肿瘤类新药的开发及临床联合用药奠定前期实验基础。Objective To carry out the pharmacodynamic study on the apoptosis of HepG2 cells induced by the effective components of Fufang Muji Granules,screen out the optimal compatibility ratio of the effective components.In addition,the new compatibility compound was formed to compare the pharmacodynamics difference between the original compound and the new compatibility compound,so as to comprehensively analyze the compatibility rule of the effective components of Fufang Muji Granules in inducing the apoptosis of HepG2 cells,and elucidate the pharmacodynamic material basis of Fufang Muji Granules.Methods Taking the effective components of Fufang Muji Granules as the study object,the microfluidic chip was designed and fabricated.Combined with the uniform design method,the apoptosis and necrosis rate was taken as the evaluation index to carry out the study on the optimal compatibility ratio of the effective components,and then a new component compatibility compound was formed.Results After treatment for 48 h,there were significant differences in apoptosis and necrosis rate of Fufang Muji Granules in the compatibility group compared with the blank group(P<0.05).The optimum compatibility ratio of pharmacodynamic components,namely,flavonoids of Juglans mandshurica Maxim,flavonoids of Cuscuta chinensis Lam,alkaloid of Sophora tonkinensis Gagnep,polysaccharide of Cuscuta chinensis Lam,polysaccharide of Sophora tonkinensis Gagnep,polysaccharide of Coriolus versicolor(L.ex Fr.)Quel was 4.35∶5.80∶15.03∶10.73∶1.36∶1.20.And through the pharmacodynamic difference comparison test,it was proved that there was no significant difference between the two efficacy results at the same dose.Conclusion By combining with microfluidic chip technology,this study preliminarily reveals the composition rule of anti-liver tumor components of Fufang Muji Granules,follow the formulation concept of traditional Chinese medicine,and lays the preliminary experimental foundation for the development of new anti-tumor drugs and clinical combined dru
