摘  要:目的:探讨北五味子提取物(SCE)对失眠大鼠模型海马组织内GABA_ARα1、GABA_ARγ2、NKCC1表达的影响。方法:制作失眠模型大鼠60只,采用随机数字表法随机分为低剂量组、中剂量组、高剂量组和模型组,每组15只,造模成功分别给予SCE100 mg/kg、200 mg/kg、400 mg/kg及10 mL/kg 0.5%CMC灌胃给药4周。并以大鼠15只为对照组。比较5组大鼠的睡眠持续时间、自主活动、海马组织内GABA_ARα1、GABA_ARγ2、NKCC1蛋白、mRNA表达水平以及行为信息。结果:失眠可增加大鼠自主活动次数、自主活动时间。SCE干预给药可减少失眠大鼠自主活动情况,改善其睡眠状况并影响行为,提高海马组织内GABA_ARα1、GABA_ARγ2蛋白、mRNA表达水平和降低海马组织内NKCC1蛋白、mRNA表达水平,且呈现一定程度的剂量依赖性(P<0.05)。结论:SCE可通过调控失眠大鼠模型海马组织内GABA_ARα1、GABA_ARγ2、NKCC1表达而改善睡眠和自主活动。Objective:To investigate the effects of Fructus Schisandrae Chinensis extract(SCE)on the expression of GABA_ARα1,GABA_ARγ2 and NKCC1 in hippocampal tissues of rats with insomnia.Methods:Sixty insomnia model rats were randomly divided into low-dose group,medium-dose group,high-dose group and model group with 15 rats in each group.SCE was given 100,200,400 mg/kg and 10 mL/kg 0.5%CMC intragastrically for 4 weeks,respectively.Fifteen rats were used as control group.Sleep duration,autonomic activity,protein and mRNA expression levels of GABA_ARα1,GABA_ARγ2 and NKCC1 in hippocampus of rats were compared in 5 groups.Results:Insomnia can increase the number and time of voluntary activity in rats.SCE intervention could reduce the autonomous activity,improve the sleep status and behavior,increase the protein and mRNA expression levels of GABA_ARα1 and GABA_ARγ2 in the hippocampus,and decrease the protein and mRNA expression levels of NKCC1 in the hippocampus,in a dose-dependent manner(P<0.05).Conclusion:SCE can improve sleep,behavior and autonomic activity by regulating the expression of GABA_ARα1,GABA_ARγ2 and NKCC1 in the hippocampus of rats with insomnia.
