摘  要:目的基于网络药理学分析黄精干预认知障碍的可能机制。方法利用中药系统药理学数据库与分析平台(TCMSP)筛选黄精的有效化合物,通过TCMSP及SwissTargetPrediction数据库筛选有效化合物靶点。通过OMIM、GeneCards、TTD、DrugBank数据库获得认知障碍的疾病相关潜在靶点。使用Venny 2.1平台获取黄精与认知障碍疾病的共同靶点。针对共同靶点,构建蛋白质与蛋白质相互作用(PPI)网络并进行可视化分析,同时进行基因本体论功能富集分析及京都基因和基因组百科全书通路富集分析。构建黄精有效化合物与疾病靶点的Circos关系图。结果本研究共筛选出黄精有效化合物12个及其靶点141个,与1880个疾病靶点取交集后获得66个共同靶点;黄精有效化合物与疾病靶点的Circos关系图提示5,4′-二羟基黄酮、黄芩素、薯蓣皂苷元、β-谷甾醇4种有效化合物募集到的靶点较多。黄精干预认知障碍的关键靶点为蛋白激酶B、β淀粉样前体蛋白、哺乳动物雷帕霉素靶蛋白、血管内皮生长因子、丝裂原活化蛋白激酶(MAPK)14等。黄精干预认知障碍的潜在靶点涉及多个细胞组分、分子功能和生物学过程,其中生物学过程主要包括细胞对氮化合物的反应、MAPK级联反应调控、老化、血液循环、神经元死亡等;同时涉及多条信号通路,主要包括神经活性配体-受体相互作用、5-羟色胺通路、磷脂酰肌醇-3-激酶-蛋白激酶B信号通路、内分泌抵抗通路等。结论黄精中的多种化合物可能通过调节多个靶点以调控MAPK级联反应调控、老化、血液循环、神经元死亡等生物学过程,以及神经活性配体-受体相互作用、5-羟色胺、磷脂酰肌醇-3-激酶-蛋白激酶B、内分泌抵抗等多个通路,从而发挥改善认知功能的作用。Objective To analyze the possible mechanism of Rhizoma Polygonati intervention on cognitive impairment based on network pharmacology.Methods The effective compounds of Rhizoma Polygonati were selected using the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and the targets of the effective compounds were screened from TCMSP and SwissTargetPrediction database.The potential targets related to the conditions of cognitive impairment were obtained through OMIM,GeneCards,TTD,and DrugBank databases.Venny 2.1 platform was used to obtain the common targets of Rhizoma Polygonati and cognitive disorders.In light of the common targets,a protein-protein interaction(PPI)network was established and a visualization analysis was performed.In addition,Gene ontology functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis were conducted on the common targets.The Circos plot for the relationship between active compounds of Rhizoma Polygonati and disease targets was developed.Results Twelve effective compounds of Rhizoma Polygonati and 141 targets thereof were identified in this study,and 66 common targets were obtained by the intersection between the targets of effective compounds and 1880 disease targets.The Circos plot for the relationship between effective compounds of Rhizoma Polygonati and disease targets indicated that four effective compounds of Rhizoma Polygonati,5,4′-dihydroxyflavone,baicalein,diosgenin andβ-sitosterol,had recruited more targets.The key targets for Rhizoma Polygonati intervention on cognitive impairment were protein kinase B,βamyloid precursor protein,mammalian target of rapamycin,vascular endothelial growth factor,mitogen-activated protein kinase(MAPK)14,etc.The potential targets for Rhizoma Polygonati intervention on cognitive impairment involved multiple cellular components,molecular functions,and biological processes,wherein the major biological processes included cellular reactions to nitrogen compound,MAPK casca
