摘 要:目的:油茶炭疽病是一种重要病害,引起油茶树落果和落蕾,落果率在20%~40%,严重时可达80%以上,给茶农带来的经济损失较大。为明确引起该病害的主要病原菌,特进行了病原菌的分离与鉴定。方法:以油茶病叶作为材料,根据科赫法则采用离体组织分离法进行分离,采用形态学结合ITS、ACT、CAL多基因系统分析鉴定。结果:从病叶中分离得到5株病原菌株DH1、DH2、DH3、DH4、DH5,根据病原菌株的形态观察和致病性测定,鉴定为炭疽菌属。PDA培养基上分别挑取菌丝体并提取基因组DNA,进行ITS、ACT、CAL PCR多基因序列联合分析,MEGA 7.0构建系统发育树,结果显示,5个菌株均为炭疽属,其中DH1、DH2、DH4、DH5为果生炭疽菌,DH3为山茶炭疽菌,形态学鉴定与分子生物学对病原菌的鉴定结果一致。结论:引起油茶炭疽病的主要病原菌为果生炭疽菌和山茶炭疽菌。Objective:Camellia oleifera anthracnose is major disease in the producing areas,which can generally cause fruit drop and bud drop.The fruit drop rate is within 20%to 40%,seriously more than 80%,causing great eco⁃nomic losses to the farmers.In order to confirm the infectious pathogen,the characteristics of the isolated pathogen were studied and identified.Method:We obtain leaves infected by C.oleifera anthracnose as research materials,iso⁃late pathogenic bacteria according to Koch,s postulates,then identified by morphological method and phylogenetic analysis based on ITS、ACT、CAL.Result:5 pathogen strains DH1、DH2、DH3、DH4、DH5 were identified based on cultural characteristics and pathogenecity of the isolates.DNA mycelium was extracted from PDA medium,ITS、ACT、CAL were amplified respectively after passing the test.The phylogenetic tree was constructed by mega7.0.The results showed that DH1、DH2、DH4、DH5 were Colletotrichum fructicola,DH3 was C.camelliae,which proved that the morphological identification was consistent with the molecular biological identification of pathogenic bacteria.Conclusion:The major pathogen were C.fructicola and C.camelliae.
