摘 要:利用重组鸡粒细胞-巨噬细胞集落刺激因子(chGM-CSF)和重组鸡白细胞介素4(chIL-4)体外诱导鸡骨髓细胞分化为鸡骨髓源树突状细胞(chicken bone marrow-derived dendritic cells,chBMDCs),对诱导条件进行优化,利用鸡巨噬细胞系HD11来源的NDV Ex刺激未成熟chBMDCs,通过形态学观察及表型鉴定来鉴定体外诱导培养所得的chBMDCs,检测chBMDCs活化情况及细胞因子水平,评估其对chBMDCs的影响,探究NDV Ex对chBMDCs生物学功能的影响。结果显示,利用protein A/G磁珠联合禽CD63抗体成功分离得到不含NDV粒子的NDV Ex,有效优化了chBMDCs的分离培养条件;NDV Ex能够降低chBMDCs MHCⅡ、CD40等表面标志物的表达,抑制chBMDCs活化。Recombinant chicken granulocyte-macrophage colony stimulating factor(chGM-CSF)and recombinant chicken interleukin 4(chIL-4)were used to induce chicken bone marrow cells to differentiate into chicken bone marrow-derived dendritic cells(chBMDCs).The induction conditions were optimized,and immature chBMDCs were stimulated with NDV Ex derived from HD11.chBMDCs obtained from in vitro induction culture were identified by morphological observation and phenotypic identification.The activation of chBMDCs and cytokine levels were examined to investigate the effect of NDV Ex on the biological function of chBMDCs.It was found that NDV Ex without NDV particles was successfully isolated using protein A/G magnetic beads combined with avian CD63 antibody,which effectively optimized the isolation and culture conditions of chBMDCs;NDV Ex could reduce the expression of surface markers such as MHCⅡand CD40 of chBMDCs and inhibit the activation of chBMDCs.This study lays the foundation for elucidating the pathogenic mechanism of NDV.
