摘  要:通过转录组学测序技术,分析LPS诱导的奶牛蹄真皮细胞基因表达谱变化,筛选差异基因并进行生物信息学分析,为从细胞和分子水平上研究奶牛蹄叶炎提供必要的依据。结果显示,10 mg/L LPS作用24 h是诱导细胞模型的最佳条件。转录组测序结果显示,LPS作用前后,共有2194个基因的表达水平发生变化,其中上调基因1138个,下调基因1056个。经过GO功能富集分析和KEGG代谢通路富集分析,发现差异基因主要富集于TNF、PI3K-Akt和MAPK信号通路等与免疫反应、炎症反应相关的通路中,推测LPS主要通过激活蹄真皮细胞相关炎性通路,诱导炎症介质的产生,从而导致奶牛蹄真皮细胞炎性损伤。Transcriptome sequencing technology was used to analyze the changes of gene expression profiles in LPS-induced hoof dermal cells,the differential genes were screened for bioinformatics analysis,so as to provide necessary basis for the future study of dairy cow laminitis at the cellular and molecular levels.The results showed that treatment with 10 mg/L LPS for 24 h was the optimal condition for the establishment of hoof dermal cell inflammatory model.The results of transcriptome sequencing showed that the expression levels of 2194 genes changed before and after LPS treatment,including 1138 up-regulated genes and 1056 down-regulated genes.GO function enrichment analysis and KEGG metabolic pathway enrichment analysis showed that the differential genes were mainly enriched in TNF signaling pathway,PI3K-Akt signaling pathway,MAPK signaling pathway and other pathways related to immune response and inflammation.It is speculated that LPS mainly induces the production of inflammatory mediators by activating the inflammatory-related pathways,thus leading to the inflammatory injury of hoof dermal cells.
