摘 要:选取体质量相近且健康的1日龄京红1号蛋鸡750只,随机均分为5组,每组6个重复,每个重复25只鸡,体质量组间差异不显著。Ⅰ组为空白对照组,在8~10日龄注射0.35 mL生理盐水,饲喂基础日粮。Ⅱ~Ⅴ组在8~10日龄时注射100 mg/kg环磷酰胺(Cy),每日1次,诱导肠黏膜免疫屏障损伤,之后分别在基础日粮中添加0,400,800,1600 mg/kg枣多糖提取物(JPE)。试验期为5周。结果显示,Cy可以抑制SIgA的分泌,进而造成肠黏膜免疫屏障损伤,而JPE则可以通过增加Toll样受体(TLR)4 mRNA表达量和微褶皱细胞(M细胞,UEA1蛋白与其数量呈正比)的数量来促进B细胞的活化;通过促进干扰素(IFN)-γ和肿瘤坏死因子(TGF)-β的分泌来促进B细胞分化;通过调节白介素(IL)-(2,4,5,6和10)和TGF-β的含量来促进B细胞的IgA类别转换和终末分化,进而提高IgA的含量,以此促进SIgA的分泌。由此得出,JPE可以通过影响IgA形成的各个阶段来促进IgA的产生,进而提高SIgA的分泌量,缓解肠黏膜免疫屏障损伤。In order to explore the effect of jujube polysaccharide extract(JPE)on intestinal mucosal immune barrier function of egg-type chicks,750 healthy 1-day-old Jinghong No.1 egg-type chicks with similar body mass were randomly divided into 5 groups with 6 replicates in each group and 25 chickens in each replicate.Group I was the blank control group,whichwas injected with 0.35 mL normal saline at the age of 8-10 and was fed with the basic diet.GroupⅡ-Ⅴwere injected with 100 mg/kg cyclophosphamide(Cy)respectively once a day at the age of 8-10 to induce intestinal mucosal immune barrier injury,and then 0,400,800 and 1600 mg/kg JPE were added to the basal diet.The experiment lasted for 5 weeks.The results showed that Cy could inhibit the secretion of SIgA and cause the damage of intestinal mucosal immune barrier,while JPE could promote the activation of B cells by increasing the expression of Toll-like receptor 4 mRNA and the number of micro fold cells(M cells,uea1 protein was proportional to its number);it could promote B cell differentiation by increasing the secretion of IFN-γand TGF-β;it could promote the IgA class conversion and terminal differentiation of B cells by regulating IL-IL-2,IL-4,IL-5,IL-6,IL-10 and TGF-β,and then increase the content of IgA,so as to promote the secretion of SIgA.It is concluded that JPE can promote the production of IgA by affecting the various stages of IgA formation,thereby increasing the secretion of SIgA and alleviating the damage of intestinal mucosal immune barrier.
