摘 要:在成熟液中添加10μmol/L和厚朴酚,44 h后,检测卵母细胞中Sirt3蛋白表达、SOD2的乙酰化水平、活性氧水平、线粒体膜电位及孤雌激活胚胎的发育能力。结果显示,在成熟液中加入和厚朴酚后,可显著提高热应激卵母细胞体外成熟率及其后续胚胎体外发育能力(P<0.05)。此外,在和厚朴酚作用下,卵母细胞中Sirt3蛋白的表达显著提高、SOD2的乙酰化水平显著降低;同时,卵母细胞ROS水平降低,线粒体膜电位升高。结果表明,和厚朴酚可提高Sirt3活性,调节SOD2乙酰化水平,从而提高热应激猪卵母细胞体外成熟率和胚胎发育能力。To investigate the potential effects of honokiol on the in vitro maturation and subsequent development of porcine oocyte under the heat stress,honokiol were supplemented into the maturation medium.After 44 h of maturation,the expression of Sirt3 protein,the acetylation level of SOD2,the level of reactive oxygen species,the mitochondrial membrane potential,and the developmental ability of parthenogenetic activation of embryos were detected.The results showed that the addition of 10μmol/L honokiol to the maturation medium could significantly increase the in vitro maturation rate of heat-stressed oocytes and the subsequent in vitro embryo development ability(P<0.05).Furthermore,the presence of honokiol during the oocyte maturation under the heat stress,the expression of Sirt3 protein in oocytes increased significantly,the acetylation level of SOD2 reduced significantly,and the level of ROS in oocytes reduced,and mitochondrial membrane potential increased.Taken together,our results indicate that honokiol can increase the activity of Sirt3 and regulate the level of SOD2 acetylation,thereby improving the in vitro maturation rate and embryonic development of heat-stressed pig oocytes.
