摘  要:目的:优选金麦含漱液的提取工艺。方法:采用HPLC法测定君药金银花中绿原酸含量,色谱柱:Kromasil C;(250 mm×6 mm,5μm);流动相:乙腈-0.4%磷酸(13∶87);检测波长:327 nm。采用牛津杯法测定不同供试品的抑菌圈直径。基于正交试验,采用常规理化指标(绿原酸含量、干浸膏得率)和生物效应指标(抑菌圈直径)相结合,运用层次分析法确定各指标的权重系数,综合评价不同提取条件,优选最佳提取工艺。结果:最终优选的最佳提取工艺条件为浸泡30 min,加水量为8倍,煎煮3次,每次30 min。结论:采用多层次多指标优选出的提取工艺科学、合理、稳定,可以用于金麦含漱液的提取。Objective:To optimize the extraction process of Jinmai gargle.Methods:The content of chlorogenic acid in Flos Lonicerae was determined by HPLC.The column was Kromasil C;(250 mm×6 mm,5μm);the mobile phase was acetonitrile-0.4%phosphoric acid(13∶87);the detection wavelength was 327 nm,and the diameter of inhibition zone was determined by Oxford cup method.Based on the orthogonal test with the conventional physical and chemical indexes(chlorogenic acid content,dry extract yield)and biological effect index(diameter of bacteriostatic zone)as the indexes,the weight coefficient of each index was determined by analytic hierarchy process,and different extraction conditions were comprehensively evaluated to optimize the extraction process.Results:The optimum extraction conditions were as follows:soaking for 30 min,adding 8 times of water,and decocting for 3 times with 30 min for each time.Conclusion:The optimized extraction process is scientific,reasonable and stable,which can be used for the extraction of Jinmai gargle.
