摘 要:目的:建立丹参-川芎药对中丹参素钠、绿原酸、迷迭香酸、阿魏酸、丹酚酸B、洋川芎内酯Ⅰ、阿魏酸松柏酯、洋川芎内酯A、藁本内酯及丹参酮Ⅱ_(A)10个成分的含量测定方法,考察不同配伍比例及配伍方式对丹参-川芎药对中10个主要活性成分溶出量的影响。方法:色谱柱:Agilent ZORBAX SB-C;柱(250 mm×4.6 mm,5μm);流动相:乙腈(A)-0.5%甲酸水溶液(B),梯度洗脱;检测波长:265,280,320 nm;进样量:20μl;柱温:35℃;流速:1.0 ml·min^(-1)。测定丹参-川芎药对的单煎液、单煎合并液(1∶1)以及不同配比的合煎液(3∶1,2∶1,1∶1,1∶2,1∶3)中上述10个主要活性成分的含量。结果:合煎液中各成分提取率明显大于单煎液和单煎合并液中提取率,且在丹参-川芎药对配比为1∶1时多种成分含量最高。结论:丹参-川芎(1∶1)的配伍比例以及合煎的配伍方式有利于10个成分在水煎液中的溶出。Objective:To establish a method for the simultaneous determination of ten components(sodium danshensu,chlorogenic acid,rosmarinic acid,ferulic acid,salvianolic acid B,senkyunolide I,coniferly ferulate,senkyunolide A,ligustilide and tanshinoneⅡ_(A))in drug pair Salvia Miltiorrhiza and Ligusticum chuanxiong,and to study the influence of compatibility proportions on the ten components.Methods:An Agilent ZORBAX SB-C;column(250 mm×4.6 mm,5μm)with the column temperature at 35℃was used;the mobile phase consisted of acetonitrile(A)-0.5%formic acid(B)(gradient elution)at the flow rate of 1.0 ml·min^(-1),and the detection wavelengths were set at 265,280 and 320 nm.The ten components were determined at different compatibility proportions,including single decoction,mixed single decoction(1:1)and mixed decoction(1∶1,1∶2,1∶3,2∶1 and 3∶1).Results:The extraction rate of combined decoction was highe than that of single decoction and mixed single decoction,and the content of multiple components was the highest when the ratio of Salvia Miltiorrhiza-Ligusticum chuanxiong was 1∶1.Conclusion:The mixed decoction of Salvia Miltiorrhiza-Ligusticum chuanxiong at the proportion of 1∶1 has an obvious promoting effect on the dissolution of components.
