摘  要:旨在建立一种高效液相色谱法,用于测定藏药二十五味松生等丸中乌头碱的含量.对HPLC法的色谱条件进行了优化,流动相为甲醇-0.5%的三乙胺(70∶30),色谱柱为Aglient ZORBAX Eclipse Plus C18(4.6 mm×250 mm,5μm),检测波长为235 nm,流速为1.0 mL/min,进样量为10μL,柱温为室温.方法学研究结果表明,乌头碱的浓度在0.059μg/mL~3.330μg/mL范围内具有良好的线性关系(r=0.9993),平均回收率为100.78%,RSD为1.16%.此方法操作简便、灵敏度高、结果准确可靠,可作为二十五味松生等丸中乌头碱的含量测定方法.To determine the content of aconitine in the Tibetan medicine Ershiwuwei Songshengdeng pills,an HPLC method was established.The chromatographic conditions of HPLC method were optimized:The determination was performed on an Aglient ZORBAX Eclipse Plus C18 column(4.6 mm×250 mm,5μm)with mobile phase methanol-0.5%triethylamine(70∶30).The detection wavelength was set at 235 nm and the flow rate was 1.0 mL/min.The injection sample volume was 10μL.The methodological study showed that the concentration of aconitine was linear in the range of 0.059μg/mL to 3.330μg/mL(r=0.9993).The average recovery was 100.78%with RSD of 1.16%.The method is simple,sensitive and accurate,and can be used as a method for the determination of aconitine in Ershiwuwei Songshengdeng pills.
