摘  要:为从线粒体角度了解银耳(Tremella fuciformis)主要栽培菌株的遗传多样性,从37个银耳栽培菌株中选取33、34和37号3个典型菌株进行高通量基因组测序,并进行线粒体基因组组装和分析,结果表明:3个菌株的线粒体基因组序列一致,但就线粒体基因组拷贝数而言,33号显著高于34号、37号。选取37号菌株线粒体基因组与已公布的7个野生菌株进行内含子多态性分析,发现它们内含子数量和插入位点各不相同,存在丰富的多态性。针对37号菌株特异内含子及周边序列设计8对引物,通过PCR扩增分析栽培菌株的多样性,结果表明:这8对引物在36个栽培菌株中均扩增出一条长度一致的条带,其长度与37号菌株的预测长度相同,说明36个菌株均含有与37号菌株相同的内含子。Mitochondrial DNA diversity of major cultivated strains of Tremella fuciformis was studied.Using high-throughput sequencing,three certificated cultivars of T.fuciformis,strain No.33,No.34 and No.37,were subjected to whole genome sequencing.Then their mitochondrial genome sequences were assembled and analyzed.The results showed that strain No.33,34 and 37 were identical in mitochondrial genome sequence,but the DNA copy number of strain No.33 was greater than that of strain No.34 and No.37.Analysis of mitochondrial intron polymorphism in strain No.37 and seven previously published wild type strains showed a rich diversity in number of introns and insertion positions between these strains.Using eight pairs of primers specific to the mitochondrial genome of strain No.37,PCR analyses were carried out on 36 cultivated strains.For all the primers,there was a band with the same length as predicted in strain No.37 in all 36 strains,suggesting all 36 strains contained the same intron as strain No.37.
