摘  要:目的观察夏枯草颗粒对乳腺增生模型大鼠乳腺组织B细胞淋巴瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)和增殖细胞核抗原(PCNA)蛋白表达的影响,探究夏枯草颗粒治疗乳腺增生作用机制。方法雌性育龄期SD大鼠30只,按照随机数字表法分为正常对照组(n=5)和乳腺增生模型组(n=25)。模型组序贯肌注苯甲酸雌二醇及黄体酮建立乳腺增生模型。建模后将模型组用随机数字表法分为:模型对照组、三苯氧胺组及夏枯草低、中、高剂量组,每组5只。各组分别予生理盐水、三苯氧胺1.8mg/kg,夏枯草颗粒0.5、1.0、3.0g/kg灌胃,共4周。给药结束后,留取组织标本,观察组织学特征,使用免疫组织化学评分评价Bcl-2、Bax、PCNA的表达。结果与模型对照组比较,三苯氧胺组及夏枯草颗粒低、中、高剂量组小叶腺泡平均数平均腺泡导管数明显减少[(6.93±2.74)、(10.80±6.42)、(10.47±5.46)、(8.93±5.36)比(15.93±7.68),P均<0.05],平均扩张导管数明显减少[(2.87±1.19)、(3.13±1.25)、(3.40±1.35)、(2.00±2.39)比(8.73±3.08),P均<0.05]。与正常对照组比较,模型对照组大鼠乳腺组织PCNA表达增加[(92.61±27.36)分比(64.57±27.93)分,P<0.05],Bax表达下降[(67.08±32.13)分比(87.63±25.80)分,P<0.05]。与模型对照组比较,夏枯草各剂量组PCNA表达明显减少[(47.07±31.51)分、(58.16±37.86)分、(30.60±32.36)分比(87.63±25.80)分,P均<0.05]。此外,与正常对照组比较,高剂量组Bcl-2表达下降明显[(101.91±16.82)分比(71.19±22.90)分,P<0.05]。结论夏枯草颗粒能改善大鼠乳腺增生,其作用机制可能与下调PNCA表达、调节Bcl-2及Bax表达有关。Objective To assess the effect of Prunella vulgaris granules on the expression of Bax, Bcl-2 and PCNA in hyperplasia mammary gland tissues in a rat model of mammary gland hyperplasia. Methods Thirty female rats with childbearing age were randomly divided into control group(n=5) and mammary gland hyperplasia model group(n=25). Mice model were developed by intramuscularly injecting estradiol benzoate and then progestin.Then the model mice were randomly divided into five groups: model control(fed with saline), tamoxifen treatment(1.8 mg/kg), and low(500 mg/kg), middle(1000 mg/kg), and high(3000 mg/kg) doses of Prunella vulgaris treatment(n=5 in each subgroup) via oral gavage for 4 weeks. After that, rats were sacrificed and tissues of rat mammary glands were collected for H&E staining and immunohistochemical staining for expressions of Bax, Bcl-2, and PCNA. Results Compared to that in the model control rats, the average number of the acinar gland in the lobular glands was decreased significantly in tamoxifen and low, middle and high doses of Prunella vulgaris treatment groups, i.e., 6.93±2.74, 10.80±6.42, 10.47±5.46, 8.93±5.36 vs. 15.93±7.68(all P<0.05), and the average number of the expanded glands was also significantly decreased in tamoxifen and low, middle, and high doses of Prunella vulgaris treated rats, i.e. 2.87±1.19, 3.13±1.25, 3.40±1.35, and 2.00±2.39 vs. 8.73±3.08(all P<0.05). Moreover, level of PCNA, Bcl-2, and Bax proteins was also altered, i.e., compared with normal rats, PCNA expression was increased(64.57±27.93 vs. 92.61±27.36;P<0.05), whereas Bax expression was decreased in the mammary tissues of the model control rats(87.63±25.80 vs. 67.08±32.13;P<0.05). Compared with the model control rats, PCNA expression in each dose of Prunella vulgaris-treated rats was significantly decreased(47.07 ±31.51, 58.16 ±37.86, and30.60 ±32.36 vs. 87.63 ±25.80;all P <0.05).
