摘 要:目的通过对泡状棘球蚴原头节、细粒棘球蚴原头节对小鼠肝脏的感染进行模拟并按不同时间节点观察感染灶,比较分析两种棘球蚴原头节感染小鼠肝组织病灶模型的钙化方式、钙化程度的差异,进一步验证并明确两种棘球蚴感染小鼠肝组织的病理转归的差异。方法将泡球蚴原头节和细粒棘球蚴原头节通过肝被膜下注射打入C57BL/6小鼠肝脏内,分别按30、90、180 d 3个时间节点针对被感染小鼠的肝组织制作组织切片,并对组织切片进行H-E染色、茜素红染色、BMP-2免疫组化染色后,对实验结果进行半定量检测并统计分析比较两组感染肝组织钙化的差异。结果棘球蚴感染一定时间后的肝组织病灶有钙化,且钙化主要分布于病灶内囊壁;两组棘球蚴原头节感染的肝组织病灶的钙化程度均随着感染时间的延长而增加;细粒棘球蚴原头节感染病灶组的钙化程度较泡状棘球蚴原头节感染病灶组的钙化程度高。结论两种棘球蚴感染肝组织的病灶均形成了纤维钙化囊壁,钙化主要位于内囊,细粒棘球蚴感染病灶的钙化程度较泡状棘球蚴感染病灶的钙化程度更高,更易形成完整的内囊壁钙化,这间接反映了两种棘球蚴在肝组织内侵袭方式和转归的不同。Objective By simulating the infection of Echinococcus multilocularis and Echinococcus granulosus on the liver of mice,observing the infection focus at different time points,we compared and analyzed the difference of calcification mode and calcification degree between the two kinds of Echinococcus protoscolex infected mice,in order to further verify and clarify the difference of pathological changes in liver tissue of mice infected with echinococcosis.Methods The protoscolex of Echinococcus granulosus and Echinococcus multilocularis were injected into the liver of C57BL/6 mice under the liver capsule.The liver tissue sections of the infected mice were made at 30 days,90 days and 180 days respectively.After H-E staining,alizarin red staining and BMP-2 immunohistochemical staining,the experimental results were semi quantitatively detected and statistically analyzed the difference of calcification in infected liver tissue between the two groups.Results Calcification was found in the liver tissue after echinococcal infection for a certain period of time,and the calcification was mainly distributed in the inner wall of the lesion;The degree of calcification of hepatic lesions in the two groups increased with the extension of infection time;The degree of calcification in Echinococcus granulosus group was higher than that in Echinococcus alveolar group.Conclusions The lesions infected with Echinococcus granulosus had a higher degree of calcification than those infected with Echinococcus granulosus,which indirectly reflected the different invasion and outcome of the two Echinococcus species in liver tissue.
