基于正交试验法的金铁锁愈伤组织培养基优化分析

摘  要:应用正交试验法对金铁锁愈伤组织诱导与芽分化离体快繁培养基进行优化。针对温度、6-BA、2,4-D、蔗糖等关键因子,用L_(9)(3^(4))表安排4因素3水平的正交试验。经方差分析后得出6-BA(1.5 mg/L)+2,4-D(0.5 mg/L)+蔗糖(30 g/L)+温度(21℃)是诱导愈伤组织的较优的培养基。光照1600 lx、光周期12 h最为优化组织培养条件。The culture medium of callus induction and bud differentiation in vitro rapid propagation was optimized by orthogonal test.According to the key factors such as temperature,6-BA,2,4-D and sucrose,the orthogonal test of 4 factors and 3 levels was arranged with L_(9)(3^(4)).The results showed that 6-BA(1.5 mg/L)+2,4-D(0.5 mg/L)+sucrose(30 g/L)+temperature(21℃)was the optimal medium for callus induction.Light 1600 lx and photoperiod 12 h were the most optimized tissue culture conditions.
