摘  要:目的:采用显微红外成像光谱对不同产地多花黄精和不同种黄精进行鉴别。方法:在波数为1018 cm^(–1)的条件下,使用显微红外成像光谱对7个不同产地多花黄精和5种黄精属植物的切片进行扫描,以糖类含量高低及分布情况作为鉴别的判断依据。结果:不同产地及不同种黄精显微图像有明显区别,表明其化学成分存在差异。不同产地多花黄精中,贵州省铜仁市样品中糖类成分含量较高,其次是黄山市祁门县和湖北省赤壁县样品,浙江省天目山、湖南省怀化市、江西省瑞昌市样品中糖类成分含量相对较低,贵州省六盘水市样品中糖类成分含量最低;5种黄精属药材中多糖类成分由高到低的顺序为黄精、长梗黄精、轮叶黄精、滇黄精、多花黄精。结论:采用显微红外成像光谱技术能够有效区分不同产地多花黄精和不同种黄精。与传统红外光谱相比,显微红外成像光谱可研究各扫描微区的组分分布情况,使其含量分析表达更加直观,具有可视性、高灵敏度等特点。Objective:To identify Polygonati Rhizoma derived from Polygonatum cyrtonema Hua of different habitats and derived from different Polygonatum species by Fourier-transform infrared microspectroscopy.Method:At the wavenumber of 1018 cm^(-1),Fourier-transform infrared microspectroscopy was employed to scan sections of Polygonati Rhizoma derived from P.cyrtonema of 7 habitats and derived from 5 Polygonatum species,and the identification was performed based on the content and distribution of polysaccharides.Results:Polygonati Rhizoma derived from P.cyrtonema of different habitats and derived from different Polygonatum species showed obvious differences in the microscopic images,suggesting the difference in the content of chemical components.Among Polygonati Rhizoma samples derived from P.cyrtonema of different habitats,the sample from Tongren City of Guizhou Province had the highest polysaccharide content,followed by the samples from Qimen County of Huangshan City and Chibi County of Hubei Province,and coming in third were samples from Tianmu Mountain of Zhejiang Province,Huaihua City of Hunan Province,and Ruichang City of Jiangxi Province.Samples from Liupanshui City of Guizhou Province had the lowest polysaccharide content.As for the samples derived from 5 Polygonatum species,polysaccharide content was in the descending order of P.sibiricum Red>P.filipesMerr>P.verticillatum(L.)Al>P.kingianum Coll.et Hemsl>P.cyrtonema.Conclusion:Polygonati Rhizoma derived from Polygonatum cyrtonema Hua of different habitats and derived from different Polygonatum species can be effectively identify by Fourier-transform infrared microspectroscopy.Compared with the conventional infrared spectroscopy,infrared microspectroscopy can be used to reveal the distribution of components in each microregion,which is more intuitive and sensitive.
