摘  要:目的观察黄芪多糖—壳聚糖/胶原(APS-CS/COL)组织工程支架在糖尿病大鼠皮肤缺损创面修复中的应用效果。方法使用壳聚糖(CS)、胶原(COL)、黄芪多糖(APS)分别制备CS/COL组织工程支架和APS-CS/COL组织工程支架,用于糖尿病皮肤缺损模型大鼠的创面修复,分别记为CS/COL组和APS-CS/COL组,每组15只,以不使用组织工程支架进行创面修复的15只糖尿病皮肤缺损模型大鼠为对照组。术后第7、14、21天,肉眼观察创面部位的大体情况,并使用ImageJ软件计算创面愈合率;取大鼠创面新生组织,HE染色后在显微镜下观察大鼠再生皮肤组织形态。结果术后第7天,对照组和CS/COL组大鼠创面可观察到明显的炎症现象,镜下见少量成纤维细胞;APS-CS/COL组大鼠创面周围未见明显的炎症现象,创面中心处可见少量肉芽组织新生,镜下见较多成纤维细胞,可观察到明显的微血管形成及组织愈合迹象。术后第14天,对照组和CS/COL组大鼠创面部位有结痂存在,存在炎症反应且创面面积较大,镜下见真皮层有大量纤维细胞和胶原纤维;APS-CS/COL组创面愈合进程较快,圆心处见小范围未愈,镜下见新生真皮层中有少量成纤维细胞和大量紧致的胶原纤维,出现了处于分化阶段的皮肤附属器。术后第21天,对照组和CS/COL组大鼠创面自圆周向内见少量上皮生长,大多被肉芽组织充填,新生白色毛发稀疏,镜下见新生皮肤成纤维细胞数量仍然较多,表皮层分化不完全且较厚,可观察到新生血管、细胞附属器;APS-CS/COL组创面被肉芽组织充填,原有创面上大部分已被上皮取代,并可见较多白色新生毛发,镜下见创面新生皮肤组织结构和正常组织相似,表皮、真皮以及新生血管、细胞附属器等结构均完全形成。APS-CS/COL组大鼠术后第7、14、21天创面愈合率均高于CS/COL组和对照组(P均<0.05)。结论APS-CS/COL组织工程支架可显著促进糖尿病大鼠皮肤缺Objective To observe the effect of astragalus polysaccharide-chitosan/collagen(APS-CS/COL)tissue engineering scaffold in repairing skin defect of diabetic rats.Methods CS/COL tissue engineering scaffolds and APS-CS/COL tissue engineering scaffolds were prepared with chitosan(CS),collagen(COL)and astragalus polysaccharide(APS)to repair the wounds of diabetic skin defects in model rats,and they were divided into the CS/COL group(n=15)and APS-CS/COL group(n=15).Fifteen diabetic skin defect model rats without tissue engineering scaffolds were used as the control group.On the 7th,14th and 21st days after operation,the gross situation of the wound was observed with naked eyes,and the wound healing rate was calculated by Image J software.The new tissue of rat wound was stained by HE staining and the morphology of regenerated skin was observed under microscope.Results On the 7th day after operation,obvious inflammation was observed on the wound surface of rats in the control group and CS/COL group,and a few fibroblasts were observed under microscope.In the APS-CS/COL group,there was no obvious inflammation around the wound,a small number of granulation tissues was observed in the center of the wound,as well as more fibroblasts,obvious signs of microvascular formation and tissue healing under the microscope.On the 14th day after operation,there were scabs on the wound site,with inflammatory reaction and large the wound area in the control group and CS/COL group,and there were a large number of fibroblasts and collagen fibers in the dermis.In the APS-CS/COL group,the wound healing process was faster,and a small area of unhealed wound was observed at the center of the circle.Microscopically,there were a few fibroblasts and a large number of tight collagen fibers in the new dermis,and there were skin appendages at the stage of differentiation.On the 21st day after surgery,a small amount of epithelial growth was observed in the wound surface of the control group and CS/COL group,most of which was filled with granulation tissu
