摘 要:目的对新疆毛菊苣中活性物质进行抗肿瘤活性筛查。方法采用CellTiter-Glo发光法,在人非小细胞肺癌细胞NCI-H358、人原位胰腺癌细胞BXPC-3、人胃癌细胞NCI-N87、人结肠癌细胞COLO-205、人乳腺癌细胞MDA-MB-453、MDA-MB-468、SK-BR-3、人乳腺导管癌细胞HCC1954肿瘤细胞株上检测毛菊苣中山莴苣素、山莴苣苦素、菊苣酸和绿原酸的体外抗肿瘤活性。结果菊苣酸和绿原酸对肿瘤细胞活力无明显影响,山莴苣素与山莴苣苦素分别对NCI-H358肿瘤细胞、MDA-MB-453人乳腺癌细胞、BXPC-3肿瘤细胞、NCI-N87人胃癌细胞、COLO-205人结肠癌细胞、MDA-MB-468人乳腺癌细胞、SK-BR-3人乳腺癌细胞和HCC1954人乳腺导管癌细胞活力的IC_(50)为28.3μmol/L与5.9μmol/L、6.237μmol/L与1.082μmol/L、36.1μmol/L与8.7μmol/L、14.8μmol/L与5.4μmol/L、8.3μmol/L与2.9μmol/L、5.5μmol/L与3.3μmol/L、10.0μmol/L与1.5μmol/L、12.8μmol/L与5.6μmol/L。Cisplatin组对MDA-MB-453人乳腺癌细胞活力的IC_(50)为15.94μmol/L。结论山莴苣素和山莴苣苦素具有较强的抑制肿瘤细胞增殖作用,其中对乳腺癌和结肠癌细胞的抑制作用较强,特别是山莴苣苦素对乳腺癌细胞增殖具有更强的抑制作用。Objective To screen the antitumor activity of active substances in Xinjiang Cichorum glandulosum Boiss.et Huet. Methods CellTiter-Glo luminescence assay was used in detecting the antitumor activity of Lactucin, Lactucopicrin, chicory acid and chlorogenic acid in Cichorum glandulosum Boiss.et Huet respectively by eight different cancerous cell lines(e.g. human non-small cell lung cancer cell NCI-H358, human in situ pancreatic cancer cell BXPC-3, human gastric cancer cell NCI-N87, human colon cancer cell COLO-205, human breast cancer cell MDA-MB-453, MDA-MB-468 and SK-BR-3, human breast duct cancer cell HCC1954).Results Chicory acid and chlorogenic acid had no significant effect on the viability of tumor cells;Lactucin and Lactucopicrin respectively increased the cell viability of NCI-H358 tumor cells, MDA-MB-453 human breast cancer cells, BXPC-3 tumor cells, NCI-N87 human gastric cancer cells, COLO-205 human colon cancer cells, MDA-MB-468 human breast cancer cells, SK-BR-3 breast cancer cells and HCC1954 human breast ductal carcinoma cells, IC_(50) was 28.3 μmol/L and 5.9 μmol/L;6.237μmol/L and 1.082 μmol/L;36.1 μmol/L and 8.7 μmol/L;14.8 μmol/L and 5.4 μmol/L;8.3 μmol/L and 2.9 μmol/L;5.5μmol/L and 3.3 μmol/L;10.0 μmol/L and 1.5 μmol/L;12.8 μmol/L and 5.6 μmol/L.In group Cisplatin, the IC_(50) activity of MDA-MB-453 breast cancer cells was 15.94 μmol/L. Conclusion Lactucin and Lactucopicrin had strong inhibitory effect on tumor cell proliferation, especially on breast and colon cancer cells. Besides, Lactucopicrin had stronger inhibitory effect on the proliferation of breast cancer cells.
