摘 要:肝损伤相关疾病是世界上主要死亡原因之一,减轻肝细胞的损伤是提高肝病患者生存率的必要条件。SIRT1,Sirtuin家族中重要的一员,是NAD+依赖性组蛋白脱乙酰基酶,与肝损伤的氧化应激、能量代谢密切相关。在此篇综述里,我们总结了目前关于SIRT1在肝损伤中作用的主要病理机制,包括抑制氧化应激、减轻炎症反应以及减少细胞凋亡。并且我们还探讨在急性肝损伤中,适度地上调或激活SIRT1可能对肝细胞具有保护性作用。Diseases related to liver injury are one of the main causes of death in the world.Reducing the damage to liver cells is the necessary requirement to improve the survival rate of patients with liver diseases.SIRT1,an important member of sirtuin family, is a DNA+-dependent histone deacetylase, which is closely related to oxidative stress and energy metabolism in liver injury. In this summarize, we sum up the main pathological mechanisms of SIRT1 in liver injury, including inhibition of oxidative stress, reduction of inflammatory response and reduction of apoptosis. In addition, we also discussed that moderate up-reglution or activation of SIRT1 may protect hepatocytes in acute liver injury.
