摘 要:目的评价电针对脓毒症相关性脑病(SAE)小鼠海马神经元焦亡的影响。方法雄性C57BL/6小鼠90只,8~12周龄,体重22~25 g。采用随机数字表法分为3组(n=30):假手术组(Sham组)、SAE组和电针治疗组(EA组)。采用盲肠结扎穿孔术建立脓毒症模型。EA组于术后2、24、48和72 h时电针双侧足三里穴。于术后7 d观察小鼠生存情况,术后第8~12天行Morris水迷宫实验,之后处死,取海马组织,采用Western blot法检测核苷酸结合寡聚化结构域样受体蛋白1(NLRP1)、含半胱氨酸的天冬氨酸蛋白水解酶1(caspase-1)、消皮素D(GSDMD)的表达水平。采用ELISA法检测海马活化的caspase-1含量。结果与Sham组比较,SAE组生存率降低,术后逃避潜伏期延长,目标象限活动时间缩短,穿越原平台次数减少,海马NLRP1、caspase-1、GSDMD表达上调,活化的caspase-1含量升高(P<0.05);与SAE组比较,EA组生存率升高,术后逃避潜伏期缩短,目标象限活动时间延长,穿越原平台次数增多,海马NLRP1、caspase-1、GSDMD表达下调,活化的caspase-1含量降低(P<0.05)。结论电针减轻小鼠SAE的机制可能与抑制海马神经元焦亡有关。Objective To evaluate the effect of electroacupuncture(EA)on pyroptosis of hippocampal neurons in mice with sepsis-associated encephalopathy(SAE).Methods Ninety male C57BL/6 mice,aged 8-12 weeks,weighing 22-25 g,were divided into 3 groups(n=30 each)by a random number table method:sham operation group(Sham group),SAE group and EA treatment group(EA group).Sepsis model was established by cecal ligation and puncture in anesthetized animals.Bilateral acupoints Zusanli(ST36)were stimulated at 2,24,48 and 72 h after surgery in group EA.The survival was observed on day 7 after surgery.Cognitive function was assessed by Morris water maze.Then the animals were sacrificed,and hippocampal tissues were obtained for determination of the expression of nucleotide-binding oligomerization domain-like receptor protein 1(NLRP1),cysteine-containing aspartate-specific protease 1(caspase-1),and Gasdermin D(GSDMD)in hippocampal neurons(by Western blot)and content of activated caspase-1(by enzyme-linked immunosorbent assay).Results Compared with group Sham,the survival rate was significantly decreased,postoperative escape latency was prolonged,the activity time spent in the target quadrant was shortened,the number of crossing the original platform was decreased,the expression of NLRP1,caspase-1 and GSDMD was up-regulated,and the content of activated caspase-1 was increased in group SAE(P<0.05).Compared with group SAE,the survival rate was significantly increased,postoperative escape latency was shortened,the activity time spent in the target quadrant was prolonged,the number of crossing the original platform was increased,the expression of NLRP1,caspase-1 and GSDMD was down-regulated,and the content of activated caspase-1 was decreased in group EA(P<0.05).Conclusion The mechanism by which EA reduces SAE may be related to inhibiting pyrolysis in hippocampal neurons of mice.
