摘 要:目的研究薏苡仁油对卵巢癌SKOV3细胞凋亡的作用及其机制。方法采用不同浓度薏苡仁油作用于卵巢癌SKOV3细胞,MTT法观察薏苡仁油对SKOV3细胞存活率的影响。采用Hoechst33342/PI染色、AO/EB染色、DNA Ladder检测、流式细胞仪检测薏苡仁油对SKOV3细胞凋亡的影响。Western blotting检测凋亡相关蛋白Bax、Bcl-2、Capase 3、8、9的表达情况。结果与对照组相比,400μg/mL和500μg/mL薏苡仁油浓度可在处理后24 h、48 h和72 h显著降低SKOV3细胞存活率(t值4.635~5.231,P<0.05),其作用随着时间的延长而增加。薏苡仁油低浓度50μg/mL、100μg/mL、200μg/mL作用于卵巢癌SKOV3细胞48h诱导凋亡效果较好。薏苡仁油作用于SKOV3细胞48h后,Hoechst33342/PI染色、AO/EB染色均可见典型的凋亡小体。不同浓度50μg/mL、100μg/mL、200μg/mL组薏苡仁油作用于SKOV3细胞48h后,与对照组相比较,细胞凋亡均显著增多(t值分别为2.536、3.012、4.908,P<0.05),其中200μg/mL组细胞总凋亡率高达(27.10±5.42)%。与对照组相比较,薏苡仁油浓度50μg/mL、100μg/mL、200μg/mL作用于SKOV3细胞48h后,Bax、Caspase 3、Caspase 8、Caspase 9蛋白表达均显著增高(t值2.436~5.709,P<0.05),Bcl-2蛋白表达显著下降(t值2.357~4.304,P<0.05)。结论薏苡仁油可诱导卵巢癌细胞株SKOV3凋亡,其机制可能与下调抑制凋亡蛋白Bcl-2、上调促凋亡蛋白Bax、Caspase 3、8、9的表达水平有关。薏苡仁油有可能抑制卵巢癌细胞的扩散、转移。Objective To study the effect and mechanism of Coix seed oil on apoptosis in SKOV3 ovarian cancer cells.Methods SKOV3 cells were treated with different concentrations of Coix seed oil.The effects of Coix seed oil on the survival rate of SKOV3 cell was examined by methyl thiazolyl tetrazolium assay.Hoechst33342/PI staining,AO/EB staining,DNA Ladder detection,and flow cytometry were used to detect the effect of Coix seed oil on apoptosis in SKOV3 cells.The expression levels of Bax、Bcl-2 and Caspase 3,8,9 were detected by western blotting.Results Compared with the control group,400μg/mL and 500μg/mL Coix seed oil significantly reduced the survival rate of SKOV3 cells at 24 hours,48 hours and 72 hours after treatment(t=4.635-5.231,P<0.05),and its effect increased with time.The best apoptosis was induced in 50μg/mL,100μg/mL and 200μg/mL groups for 48 hours.Typical apoptotic bodies were observed in Hoechst33342/PI staining and AO/EB staining after SKOV3 cells were treated with Coix seed oil for 48 hours.Compared with the control group,the apoptosis of SKOV3 cells was significantly increased after treating for 48 hours in the different concentrations of 50μg/mL,100μg/mL and 200μg/mL Coix seed oil groups(t=2.536,3.012 and 4.908,respectively,P<0.05).The total apoptosis rate of 200μg/mL group was as high as(27.10±5.42)%.Compared with the control group,the protein expressions of Bax,Caspase 3,Caspase 8 and Caspase 9 after treating with Coix seed oil for 48 hours in 50μg/mL,100μg/mL and 200μg/mL groups were significantly increased(t=2.436-5.709,P<0.05),and the protein expressions of Bcl-2 were significantly decreased(t=2.357-4.304,P<0.05).Conclusion Coix seed oil could induce the apoptosis of SKOV3 ovarian cancer cells,which may be related to the down-regulation the expression levels of anti-apoptotic protein Bcl-2 and up-regulation the expression levels of pro-apoptotic protein of Bax and Caspase 3,8,9.Coix seed oil may inhibit the spread and metastasis of ovarian cancer cells.
