摘  要:目的探讨膈下逐瘀汤"异病同治"子宫内膜异位症(EMs)和多囊卵巢综合征(PCOS)的分子作用机制。方法TCMSP数据库获取膈下逐瘀汤活性成分及预测药物作用靶基因,MelaCards和CTD数据库获取EMs和PCOS疾病相关靶基因,将EMs、PCOS疾病相关靶基因与膈下逐瘀汤作用靶基因取交集获取共有靶基因,STRING在线工具构建共有靶基因蛋白互作网络,Cytoscape软件进行共有靶基因蛋白网络可视化处理,clusterProfiler工具进行共有靶基因GO及KEGG信号通路分析,cytoHubba、MCODE插件筛选核心靶基因,SwissDock对核心成分与核心靶基因进行分子对接。结果膈下逐瘀汤治疗EMs、PCOS的共有靶基因共69个,GO功能涉及激素合成与反应、炎症等方面,KEGG信号通路结果显示交集基因富集于与炎症相关的IL-17、TNF、Toll样受体信号传导途径和Th17细胞分化,与激素相关的内分泌抵抗、卵巢类固醇生成、类固醇激素的生物合成、甲状腺激素、雌激素等信号转导通路。共有靶基因PPI网络中排名前3位的核心基因为IL-6、IL-10和JUN,PPI网络中计算出核心子网络共4个,均与炎症、血管生成和激素合成密切相关。结论膈下逐瘀汤治疗EMs、PCOS的作用机制与调节激素水平、反应及调控炎症和调节血管生成等密切相关。Objective To explore the molecular mechanism of Gexia Zhuyu decoction for“treating different diseases with same method”in the treatment of endometriosis(EMs)and polycystic ovary syndrome(PCOS).Methods The active components of Gexia Zhuyu decoction and predicting target genes of drugs were obtained from TCMSP database,and EMs-and PCOS-related disease target genes were obtained from MelaCards and CTD database.The EMs-and PCOS-related target genes were intersected with the Gexia Zhuyu Decoction target genes to obtain the common target genes.The STRING online tool was used to construct the protein interaction network of common target genes,the Cytoscape software was used to visualize the protein network of common target genes,the clusterProfiler was used for GO and KEGG signal pathway analysis of common target genes,and the cytoHubba and MCODE plug-ins were used to find out the core target genes.Molecular docking of core components and core target genes was conducted by SwissDock.Results A total of 69 common target genes were shared in the process of Gexia Zhuyu decoction in treating EMs and PCOS.GO functions involved in the hormone synthesis and response,inflammation,etc.The KEGG signalling pathway results showed an enrichment of intersecting genes in inflammation-related IL-17,TNF,Toll-like receptor signalling pathways,Th17 cell differentiation,hormone-related endocrine resistance,ovarian steroidogenesis,steroid hormone biosynthesis,thyroid hormone,oestrogen and other signalling pathways.The top 3 core genes in the PPI network of the shared target genes were IL-6,IL-10 and JUN,and there were 4 core sub-networks in the PPI network,which were all closely associated with inflammation,angiogenesis,and hormone synthesis.Conclusion The mechanism of Gexia Zhuyu decoction for"treating different diseases with same method"in the treatment of EMs and PCOS is closely related to the regulation of hormone levels and responses,modulation of inflammation,and regulation of angiogenesis.
