发酵彝药对小鼠肠道菌群的重建作用

摘  要:目的研究发酵彝药解本10号和303号对盐酸林可霉素致小鼠肠道菌群失调的重建作用。方法采用盐酸林可霉素建立小鼠肠道菌群失调模型,按12 mL·kg^(-1)的剂量灌胃给予小鼠不同浓度的解本10号和303号,连续7 d。同时设正常对照组、自然恢复组。实验结束,取小鼠回盲肠的粪便,提取DNA,进行16S rRNA高通量测序,并进行生物信息学分析。结果解本10号和303号可显著提高菌群失调小鼠肠道内的菌群种类和丰度,使有害菌减少,益生菌增多。结论发酵彝药解本10号和303号可有效重建失调的肠道菌群。OBJECTIVE To investigate effects of fermented Yi medicine JIEBEN 10(JB10)and JIEBEN 303(JB303)on re-establishment of gut microbiota in mice with dysbacteriosis induced by using lincomycin hydrochloride.METHODS Mouse model of intestinal dysbacteriosis was established by using lincomycin hydrochloride.Then the mice were intragastrically administrated with different concentrations of JB10 and JB303 with the doses of 12 mL·kg^(-1) for 7 days,while normal group and natural recovery group were set up.After completion of the experiment,fecal samples in mice ileocecum were collected and the total DNA were extracted and analyzed with 16 S rRNA high throughput sequencing,and then the data were analyzed by using bioinformatics.RESULTS The diversity and abundance of gut microbiota in mice with dysbacteriosis were significantly increased by JB10 and JB303,while the abundance of harmful bacteria were decreased and probiotics were significantly increased.CONCLUSION Fermented Yi medicine JB10 and JB303 can effectively re-establish the imbalanced gut microbiota.
