摘  要:目的:建立2型糖尿病波动高血糖模型,为波动高血糖的相关研究奠定实验基础。方法:选取4~6周龄雄性Wistar大鼠(n=40),适应性喂养1周,将其随机分为对照组(Control,C,n=10)、2型糖尿病组(Type 2 Diabetes Mellitus,T2DM,n=30)。T2DM组大鼠高糖高脂饲料4周(High Glucose and Lipid)并联合小剂量链脲佐菌素(Streptozotocin,STZ),诱导2型糖尿病模型。将成功的2型糖尿病组大鼠(n=27)随机分为持续高糖组(Constant high glucose group,CHG,n=13)及波动高糖组(Intermittent high glucose group,IHG,n=14),IHG组通过改变大鼠生活节律和饮食习惯来建立。观察各组大鼠一般情况、大鼠日间的代谢情况及日间血糖波动情况。结果:一般状况:C组较好,CHG组次之,IHG组最差;死亡率:CHG组38.5%,IHG组42.9%。体质量:第1周:三组间体质量差异无统计学意义(P>0.05);第5周:与C组相比,CHG组及IHG组体质量均显著增高(P<0.05);第11周:体质量C组与CHG组均显著高于IHG组(P<0.05);第16周:体质量C组最高,CHG组居中,IHG组最低(P<0.05)。代谢量:进食:C组与IHG组均显著少于CHG组(P<0.05);饮水量及尿量:CHG组最高,IHG组居中,C组最低(P<0.05)。日间9个时间点血糖:组间比较、时间点比较及交互作用差异均有统计学意义(P<0.05),提示不同组间、不同时间点血糖水平有显著差异,并且组间与时间存在显著的交互作用。结论:高糖高脂联合小剂量STZ的基础上,通过改变大鼠饮食习惯和生活节律可以成功建立波动性高血糖模型。Objective:To establish a model of intermittent high glucose in type 2 diabetes mellitus(T2DM),and to lay an experimental foundation for the related research of fluctuating hyperglycemia.Methods:Male Wistar rats aged 4-6 weeks(n=40)were randomly divided into control group(C,n=10)and type 2 diabetes mellitus group(T2DM,n=30)after adaptive feeding for one week.T2DM rats were fed with high glucose and high fat diet for 4 weeks combined with low-dose streptozotocin(STZ)to induce T2DM model.Successful type 2 diabetic rats(n=27)were randomly divided into constant high glucose group(CHG,n=13)and intermittent high glucose group(IHG,n=14).IHG group was established by changing the life rhythm and eating habits of rats.The general condition,daytime metabolism and blood glucose fluctuation of rats in each group were observed.Results:General condition:Group C was better,followed by CHG,IHG was the worst.Mortality:CHG group was 38.5%,IHG group was 42.9%.Body weight:Week 1,there was no significant difference among the three groups(P>0.05);week 5,compared with group C,the body mass of CHG group and IHG group were significantly increased(P<0.05);week 11,the body mass of C group and CHG group were significantly higher than that of IHG group(P<0.05);week 16,the body mass of group C was the highest,CHG group was the middle,and IHG group was the lowest(P<0.05).Metabolism:food intake:group C and IHG group were significantly less than CHG group(P<0.05);water consumption and urine volume:CHG group was the highest,IHG group was the middle,group C was the lowest(P<0.05).Blood glucose at 9 time points during the day:there were significant differences in the comparison among groups,time points and interaction(P<0.05),it suggested that there was a significant differences in blood glucose levels between non groups and at different time points,and there was a significant interaction between groups and time.Conclusion:On the basis of high glucose and high fat combined with small dose of STZ,the rat model of intermittent high glucose can be succe
